In this study,carbon tetrachloride was used to induce acute liver injury in mice.The preventive and therapeutic effects of camel urine on liver were studied through the analysis of serum biochemical indicators and histopathology,as well as the analysis of antioxidant,inflammatory factors and fluorescence quantitative PCR in liver tissues of experimental mice.Through the liver injury prevention experiment,it was found that,compared with the model group,the activities of ALT,AST,TBIL,ALP and GLU in serum of the camel urine group with low(170 mg·kg-1),medium(340 mg· kg-1)and high(500 mg·kg-1)doses were significantly reduced,while the activities of ALB and TP were increased.The observation of pathological sections of mouse liver showed that the liver injury was improved in different degrees in camel urine groups.Compared with the model group,serum AST,ALT,ALP,UA,TNF-a and IL-6 activity in the camel group were significantly decreased after treatment of liver injury(500 mg·kg-1)and(670 mg·kg-1)doses(p<0.05).MDA and GSH levels in liver tissues of mice in the low and high dose camel urine group and the positive control group were significantly lower than those in the model group(p<0.05).Compared with the model group,the liver histological injury was significantly reduced in the low and high dose camel urine group and the positive control group.Fluorescence quantitative PCR results showed that COX-2 gene was significantly up-regulated in the model group,while CYP2el gene was down-regulated,which was statistically significant compared with the normal group and the camel urine low-dose group.The inflammatory genes IL-1? and TNF-? were down-regulated in the low and high dose group of camel urine,while the IL-6 gene was significantly up-regulated in the model group,which was significantly different from the normal group and the low and high dose group of camel urine.Studies have found that camel urine has the ability to scavenge DPPH free radicals,iron ion chelating ability,hydroxyl free radical scavenging ability,and hydroxyl free radical scavenging ability is the strongest,so camel urine may play a protective role in liver injury prevention and treatment by scavenging hydroxyl free radical.According to the comprehensive research results,camel urine has obvious preventive and therapeutic effects on chemical liver injury,which can enhance the antioxidant capacity of the liver,reduce the secretion of inflammatory factors,provide experimental basis for the prevention and treatment of liver injury and disease,and promote its wider application in medical treatment.
