Objective:This paper aims to study the protective effect of camel urine on alcohol-induced acute liver injury in mice,and explore the toxicology of camel urine.Methods:(1)camel urine acute toxicology experiment was to inject with camel urine freeze-dried powder at the dose of 4500 mg/kg.bw to healthy Balb/c mice Within 24 hours.and equal volume of normal saline was given to the blank control group,then observated for 7 days.(2)Subacute toxicological tests was to inject to mice with camel urine freeze-dried powder divided into three doses which was 1000 mg/kg.bw,500 mg/kg.bw and 250 mg/kg.bw for 4 weeks,10 mice in each group and the volume was 10 mL/kg.(3)Prevention and protection test of camel urine on alcoholic acute liver injury in mice:40 SPF male ICR mice were randomLy divided into 5 groups according to body weight,which were blank control group,model group,camel urine low dose group,camel urine high dose group and positive drug control group respectively.The low-dose camel urine group and the high-dose camel urine group were intragastrically administered with 250 mg/kg.bw and 500 mg/kg.bw camel urine lyophilized powder respectively.The positive drug group was given a dose of 150 mg/kg.bw silymarin through administmiceion by gavage,and the gavage volume was 10 mL/kg.The blank control group and the alcoholic liver model group were given an equal volume of normal saline.They were all given by gavage once a day for 28 days simultaneously.After 28 days,except for the blank control group,the other 4 groups were given 50%ethanol(17.7 mL/kg)to replicate the model of acute alcoholic liver disease.After fasting for 12 h,the mice were weighed and sacrificed by cervical dislocation.AST and ALT activities and levels of TNF-?,IL-1?,IL-6,and MDA,SOD,GSH-Px and TG levels in liver tissues were measured,and his topathological examination was performed on the liver.Results:(1)The result of camel urine acute toxicity experiment showed that it did not produce toxic reaction at the does of 4500mg/kg to mice within 24 hours.(2)Subacute toxicological tests showed that Blood biochemical index and the hematological indexes of mice in each period and each dose group had no significant differences with the normal group(p>0.05)there was no toxicity reaction.The main organs heart,liver,spleen,kidney of mice had no lesion.(3)The results showed that the levels of AST,ALT,TNF-?,IL-1?,IL-6 and TG were significantly increased compared with the control group(p<0.01);and the level of SOD and GSH-PX were significantly decreased(p<0.01),which means the model is successful.Compared with the model group,the levels of AST,ALT,TNF-?,IL-1?,IL-6 and TG were significantly decreased(p<0.01);and the level of SOD and GSH-PX were significantly increased(p<0.01).Conclusion:The test results show that it is safety to use camel urine in mice,and there is no obvious side effect and,the camel urine has a preventive and protective effect on alcohol-induced acute liver injury.
