ObjectivesTo study the protective effects of sedum sarmentosum on acute liver injury mice induced by carbon tetrachloride and the mechanism of action, which could provide scientific evidence for further study and clinical use.Methods1. C57BL/6mice were administrated by single intraperitoneal injection with CCl4-olive solution. Compared the aminotransferase levels in plasma of CCl4-induced liver injury mice at different doses and time points to choose a appropriate model preparing for this study.2. Screened effective formulas and herbs that can alleviate hepatitis from Yin-chen-hao Decoction, Huang-lian-jie-du Decoction, Xi-jiao-di-huang Decoction, Xiao-yao-san Decoction, Bu-zhong-yi-qi Decoction and hypericum japonicum, bupleurum, sedum sarmentosum through CCl4-induced mice model, and evaluate the curative effect by analyzing serum aminotransferase levels and liver pathology with HE staining.3. The mice with acute liver injury were induced by CCl4. By analyzing the proportions and numbers of inflammatory cells in peripheral blood and livers of the mice, detecting the secretion and expression of inflammatory cytokines and testing the indicators of oxidative stress in liver, we explored the protective mechanism of sedum sarmentosum on acute liver injury.Results1. The increase of aminotransferase levels was in a dose-dependent manner with CCl4by single intraperitoneal administration.Aminotransferase levels increased at first and then declined.2. The biochemical tests showed sedum sarmentosum decreased the serum aminotransferase levels significantly. Liver pathology results (HE staining) showed that sedum sarmentosum alleviated the hepatocytes necrosis and reduced the inflammatory cell infiltration in liver of CCl4model.3. Sedum sarmentosum reduced the expression of the cytokines and factors such as TNF-α, MCP-1, IL-6and iNOS in liver tissue,and effectively reduced the level of MDA in liver. However, it did not affect the proportions and numbers of inflammatory cells in peripheral blood and liver of the mice.Conclusions1. C57BL/6mice model of mild acute liver injury can be prepared by injecting0.1%of CCl4-olive solution.Sixteen hours after injection is a suitable time point for the detection of a number of indicators.2Sedum sarmentosum is the curative herb to mice of acute liver injury induced by CC14.3. A potential protection mechanism is that sedum sarmentosum may reduce the liver damage and protect the liver by reducing lipid peroxidation caused by free radical in liver.
