Objective:The dry flower of Imperial Chrysanthemum of Compositae has many values on orna--mental,edible,health care,it has some effects on antioxygenation,antibacterial,lowering blood pressure,regulation of blood glucose.But the lack of quality evaluation method nowdays,and For conducting a reasonable evaluation of imperial chrysanthemum resources,make full use of the imperial chrysanthemum chrysanthemum resources,to further develop the emperor,This paper provides theoretical foundation though morph--ological identification,microscopical identification,Flavonoids content determination,anti--oxidant activity research of flavonoids extract,determination of active ingredients and establishment of HPLC fingerprint analysis method for 16 different areas of Imperial Chrysanthemum,aim to provides basic data and scientific basis for the quality standards standardization construction of the Imperial chrysanthemum.Methods and results:1.morphological and microscopical identification:Collected 16 different areas of the Imperial chrysanthemum samples morphological and microscopical identification study.morphological identification:The tongue of the imperial chrysanthemum flowers are yellow,tubiform floret is yellow or yellow-brown,total bracts is green or brown;micro-scopical identification:parenchyma cells,T-shaped hairs,sessile glandular hairs,nonglan--dular hairs,pollen grains,anther basal cells,base of anther cell tubiform floret pistil stigma,tubiform floret stamen anther and catheter.2.Determination of total flavonoids:With the extract of total flavonoids as evaluation index,effects of grind degrees,ultrasonic temperature,extraction time,solvent-to-solid ratio,ethanol concentration on the yield of total flavonoids by sigle factor tests and response surface methodology.Optimum ultrasound extraction process was as following:the crush--ing degreeis 80 mesh,the ultrasonic temperature is 59?,extraction time was 46min,solid to liquid ratio of 45:1,79%ethanol concentration.The content of total flavonoids from 16 different areas of the Imperial chrysanthemum was determined to be 60.45 and 146.59 mg/g.3.The antioxidant activity of total flavonoids extract from Imperial chrysanthemum:with ascorbic acid(VC),2,6-tert butyl-4-methyl phenol(BHT)as reference,and scavenging DPPH free radical ability,scavenging ABTS free radical ability,against hydroxyl free radicals and the total reducing capacity as evaluating indicator.The research showed that the antioxidant activity of total flavonoids extract from Imperial chrysanthemum was good in the experimental concentration linear range(0.15?1.00mg/ml),and S3?S8?S11 compo--site antioxidant capacity is best.4.Determinnation of various activi ingredients by HPLC:Agilent Technologies HC-C18(2)(4.6mm×250mm,5?m)was adopted,with acetonitrile-0.2%phosphoric as the mobie phase at the flow rate of 0.8ml/min,the columteperature was set at 30?,the UV detection wavelength were 348 nm and the Sample volume was 10?l,the order of Gradient elution:(0?10min,10%A?18%A;10?30min,18%A?26%A;30?55min,26%A?38%A;55?70min,38%A?60%A;70?75min,60%A).The results showed that the activi ingredients were different between the imperial chrysanthemum in different regions,and 9 kinds of combined che--mical composition content is of the highest in linyi in Shandong.5.Analysis of HPLC fingerprint of the imperial chrysanthemum:The fingerprint of the imperial chrysanthemum that were from 16 different producing areas was established using HPLC,the study found 15 common peaks and 6 common chemical composition were Chlorogenic acid,mignonette glycosides,chlorogenic acid,baicalin A,Flowers glycosides and celery.The similarity calculation were established using traditional Chinese medicine chromatographic fingerprint evaluation system(2004 version A).The similarity calculation results,ranged from 0.642?0.99 in the 16 different areas of the Imperial chrysanthemum.According to the cluster analysis and principal component analysis of 15 common peaks,the samples of Emperor Chrysanthemum were classified,and the results of cluster analysis and principal component analysis were basically consistent with the results of similarity analysis..
