Objective:To make quality evaluation and identify accurately efficiently and conveniently for Dendrobium officinale?Dendrobium nobile?Dendrobiumchrysotoxum?Dendrobium thyrsiflorum?Dendrobium wardianum?and Dendrobium aphyllum.Methods:Based on the differences of HPLC fingerprint,ultraviolet spectroscopy,super high performance liquid-mass spectrometry and statistics,six species of Dendrobium are studied in order that they can be identified.The differences of naringenin,gigantol,erianin,polysaccharides,mannose,glucose and amino acid are studied in order to evaluate the quality of Dendrobium.Results:HPLC fingerprint of six species Dendrobium are established and the similarity between 0.747 and 0.933.Totally 25 common peaks are found and based on the retention time and absorption spectra of reference compounds,three constituents including naringenin(20)gigantol(22)and erianin(24)are identified.Combined the differences of fingerprin,cluster analysis and principal component analysis,all the Dendrobiums could be differentiated.According to the differences of the shape,the maximum absorption and absorbance,six kinds of Dendrobium can be differentiated.UPLC-MS totally detected six compounds including naringenin,gigantol,erianin,chrysotoxine,moscatilin and chrysotobibenzyl.Among them,there are all detected in Dendrobium chrysotoxum.Detection of naringenin,gigantol and erianin in Dendrobium officinale.Detection of naringenin and gigantol in Dendrobium nobile.Only moscatilin is detected in Dendrobium aphyllum.chrysotoxine is not detected in Dendrobium wardianum and chrysotobibenzyl is not detected in Dendrobium thyrsiflorum.Six species of Dendrobium can be identified according to the differences of these detected compounds.Quantitative and analysis of variance results showed that the content of naringenin,gigantol,erianin,polysaccharides,mannose,glucose and amino acid exist significiantly differencein the six kinds of Dendrobium(P<0.05).The content of erianin is the highest in Dendrobium chrysotoxum and the mean value is(1353±91.67)ug/g.The contents of naringenin and gigantol are the highest in the Dendrobium thyrsiflorum,the mean values are(152±21.28)ug/g and(894± 15.82)ug/g,followed by Dendrobium aphyllum(P<0.05),the other are no significant difference(P>0.05).From the contents of naringenin gigantol and erianin,Dendrobium chrysotoxum,Dendrobium thyrsiflorum and Dendrobium aphyllum are superior to others.The contents of polysaccharides in Dendrobium officinale are the highest and peak area ratios of mannose and glucose meet Pharmacopeial regulations(2.4-8.0)(P<0.05),followed by Dendrobium aphyllum,the other are no significant difference.From this point of view,Dendrobium officinale and Dendrobium aphyllum are superior to others.The amino acid content of Dendrobium officinale,Dendrobium nobile,and Dendrobium aphyllum is higher than the others(P<0.05).Conclusion:High-performance liquid chromatography,ultraviolet spectroscopy and mass spectrometry information can identify Dendrobium.The quantitative of multi-components can evaluate the quality of Dendrobium to a certain extent.They can provide reference for identification and quality evaluation of Dendrobium.
