OBJECTIVE: To investigate the effect of Linc01510 on the invasion and migration of gastric cancer cells,and the mechanism was explored from the perspective of miR-93-5p/IFNAR1 pathway.METHODS: After up-regulating Linc01510,Transwell assay was used to evaluate the effect of Linc01510 on the invasion and migration of gastric cancer cells.Bioinformatics analysis predicted miRNAs with a targeted relationship with Linc01510;whether the interaction between Linc01510 and miR-93-5p was demonstrated by dual luciferase reporter gene experiments.The expression levels of Linc01510,miR-93-5p and IFNAR1 mRNA after up-regulation of Linc01510 were detected by real-time quantitative PCR(qRT-PCR).The expression level of IFNAR1 protein was detected by Western blot.RESULTS: Results of Transwell experiments showed that overexpression of Linc01510 could inhibit the invasion and migration of gastric cancer cells.Bioinformatics analysis predicted Linc01510-related miRNAs.The results of dual luciferase reporter assay demonstrated a direct interaction between Linc01510 and miR-93-5p.Overexpression of Linc01510 could down-regulated the expression level of miR-93-5p and up-regulated the expression level of IFNAR1(P<0.05).CONCLUSION: Linc01510 inhibits invasion and migration of gastric cancer cells,and its mechanism may be regulated by the miR-93-5p/IFNAR1 pathway.
