Objective: The objective of this study is to analyze the differentially expressed proteins in cystic wall tissues of ovarian endometriotic cyst,ovarian cyst and normal ovarian tissues.Also,the differentially expressed proteins related to the recurrence of ovarian endometriotic cyst will be identified as markers and its clinical value will be explored.Methods: Tissues of ovarian endometriosis cyst wall,benign ovarian cyst wall and normal ovary(6 specimens in each group)were collected.Differentially expressed proteins of all tissues were screened out by isobaric tags for relative and absolute quantitative(iTRAQ)and the differentially expressed proteins related to the recurrence of ovarian endometriotic cyst were identified by iTRAQ combined with functional annotation and biological information analysis.All the identified differentially expressed proteins related to the recurrence were validated using immunohistochemical method.Results : A total of 359 up-regulated differentially expressed proteins were identified in all three groups.Compared with normal ovary and benign ovarian cyst,27 high expressed proteins were found in ovarian endometriotic cyst group in condition of above 2 folds of protein difference.The top five proteins with remarkably up-regulated expression were CLC,DEFA1,S100A9,S100A8,FTL.The immunohistochemical test results showed that the variation trend of the differentially expressed proteins S100A9 and S100A8 were consistent with the results of iTRAQ detection.No similar trends were found in CLC,DEFA1 and FTL.The positive rate of high expression of the proteins S100A9 and S100A8 in the recurrent group of ovarian endometriotic cyst was higher than that in the non-recurrent group.A novel unknown function protein(A8K3Q9)was found which are highly expressed in ovarian endometriotic cysts.Conclusion: There existed differentially expressed proteins with high expression in ovarian endometriotic cysts.The identified differentially expressed proteins S100A9 and S100A8 and may be molecular markers that predict postoperative recurrence of ovarian endometriotic cyst and guide subsequent treatment.
