The Expression And Clinical Significance Of HOXA10,HOXA9,PAX8and Calretinin In Ovarian Endometriotic Cyst

Objective:To investigate the expression and the clinical significance of HOXA10,HOXA9,PAX8and Calretinin in ovarian endometriosis cyst tissues.Material and Methods:1clinical material:27cases of ovarian endometriosis cyst specimens were collected form patients of shandong qilu hospital from January2009to March2012. The specimens including12cases of fresh ovarian endometriosis cyst specimens,15cases of paraffin ovarian endometriosis cyst organization.and96cases of normal endometrium and normal fallopian tube specimens were collected, including38cases of fresh normal endometrium specimens and38cases of normal fallopian tube specimen,11cases of Paraffin normal endometrium organization,9cases of paraffin normal fallopian tube organization.these patients were preoperative suspected ovarian malignant dieases and recepted bilateral or unilateral accessories resection, postoperative confirmed were benign lesions, at the same time recepted D&C or hysterectomy, the patients with Ovarian endometriosis cyst were from23to47years old, median age was33.2years. Annex resection patients were28～54years old, the median age of40.3years. All of the patients have complete clinical and pathological material, and did not accept the preoperative hormone therapy, all of the cases were postoperative confirmed by pathological examination. 2methods(1)the expression of HOXA9and HOXA10protein in the normal endometrium and the fallopian tube organization were tested by the western-blotting and immunohistochemical methods.(2) the expression of PAX8, Calretinin, HOXA9and HOXA10in the ovarian endometriosis cyst tissues were tested by the western-blotting and immunohistochemical methods, in order to detecting the origin source of ovarian endometriosis cyst3statistical methodsThe SPSS13.0statistical analysis software was used to the comparison between different groups with t test,the comparison between more than two groups were measured with the single factor analysis of variance.Results:1The expression and significance of HOXA9and HOXA10in normal endometrium and fallopian tubal tissue1.1The HOXA9and HOXA10expressed in all the specimens of normal endometrium tissues, in the epithelial cell cytoplasm of the endometrial glands, No significant differences between the HOXA9and HOXA10expression(p>0.05);The HOXA9and HOXA10expressed in all the specimens of normal fallopian tube tissues, in the epithelial cell cytoplasm of the fallopian tubal mucosa epithelium, the expression of HOXA9was higher than HOXA10,but with no significant differences between the two groups(p>0.05);1.2Western-blotting detection shows that HOXA9expressed in both endometrium and fallopian tubal umbrella tissues, the expression in the fallopian tubal umbrella tissues were higher, with significant statistical difference (p<0.05);HOXA10expressed in both endometrium and fallopian tubal umbrella tissues, but with no significant statistical difference (p>0.05). 2The expression and significance of PAX8, Calretinin, HOXA9and HOXA10in ovarian endometriosis cyst tissue.2.1Western-blotting results shows that PAX8, Calretinin, HOXA9and HOXA10expressed in all of the ovarian endometriotic cyst tissues.2.2The immunohistochemical staining of PAX8, Calretinin, HOXA9and HOXA10shows that there are endometrial glands-like structures in ovarian endometriosis cysts tissuss,they are dense layer of cells, cells are polygonal, with clear borders, closely spaced, and with granular cytoplasm, nuclear is big and round, nucleoli are obvious. Immunohistochemical stainning shows:(1) PAX8expressed in the endometrial glands-like structures of15cases of ovarian endometriosis cyst samples,positive location were in the nuclear of epithelial cells;(2) HOXA10expressed in the endometrial glands-like structures of15cases of ovarian endometriosis cyst samples,positive location were in the cytoplasm of epithelial cells;(3) HOXA9expressed in the endometrial glands-like structures of5cases of ovarian endometriosis cyst samples,positive location were in the cytoplasm of epithelial cells;but were negatively expressed in the endometrial glands-like structures of10cases of ovarian endometriosis cyst samples;(4) Calretinin expressed in15cases of ovarian endometriosis cyst samples, the positive location were in the nuclear of ovarian germinal epithelial cells and stroma, but were negatively expressed in the endometrial glands-like structures of all cases of ovarian endometriosis cyst samplesConclusions:1HOXA9and HOXA10are related to the development of endometrium and fallopian tube, in which the HOXA9is strong correlate to the development of the Fallopian tube. 2The ovarian endometriosis cyst may origin from the endometrium tissue.
