| Lepidium latifolium L.is a perennial herb of Lepidium of Brassicaceae.Based on the study of promoting gastrointestinal motility in the early stage,this paper further studied the mechanism of action of medicinal ingredients of promoting gastrointestinal motility.To lay the foundation for its follow-up research and development.In this study,the aqueous extract(TAE)of L.latifolium L.was used as the test drug,and SD rats were used as experimental animals and its effects of gastrointestinal hormones,Cx43 and C-Kit were investigated at the tissue level.At the same time,the effects of different components in the extract of L.latifolium L.on the contraction and proliferation of rat gastrointestinal smooth muscle cells were observed by microscopic micrometric techniques and CCK-8 method.Furthermore,the effects of SP,Ghrelin,VIP,Cx43 mRNA,P38 MAPK mRNA,PKA mRNA,PKC mRNA and M2R mRNA were investigated at the cellular and molecular levels,and the mechanism of action was also explored.The research results are as follows:(1)TAE of L.latifolium L.significantly increased SP and Ghrelin content in the stomach and duodenum,but had no effect on gastric and duodenal VIP content.(2)The results of immunohistochemistry showed that TAE of L.latifolium L.was not significant for the expression of positive Cx43 and C-Kit.(3)Different components of extracts from L.latifolium L.could promote cell contraction and increase cell contraction rate,but had no significant effect on cell proliferation.(4)Component V and the part of ethyl acetate(PEA)can increase the content of SP and Ghrelin in the culture supernatant of rat gastric smooth muscle cell,component V can decrease VIP content;component II and PEA can increase the content of Ghrelin in the culture supernatant of rat small intestinal smooth muscle cells.(5)Component V can increase the relative expression of PKA mRNA in rat gastric smooth muscle cells.PEA could increase the relative expression of M2R mRNA in gastric smooth muscle cells of rats.PEA can increase the relative expression of Cx43 mRNA,P38 MAPK mRNA,PKA mRNA,PKC mRNA and M2R mRNA in rat small intestinal smooth muscle cells.Component VII could increase the relative expression of PKA mRNA in rat colonic smooth muscle cells,and PEA could increase the relative expression of Cx43 mRNA and M2R mRNA in rat colonic smooth muscle cells.In summary,the medicinal ingredients of promoting gastrointestinal motility of L.latifolium L.play a role mainly by stimulating the contraction of gastrointestinal smooth muscle cells.Its promoting gastrointestinal motility mechanism is associated with elevating SP and Ghrelin content,up-regulation of Cx43 mRNA,P38 MAPK mRNA,PKA mRNA,PKC mRNA and M2R mRNA expression. |
