Lepidium latifolium L is a speicies of perennial herb belonging to Lepidium apetalum of Cruciferae family.The previous study has shown that the ethyl acetate extract can improve the gastrointestinal motility.To futher pick up the effective substance,we separated the ethyl acetate from Lepidium latifolium L using the tech of slica column chromatography,and went on testing the pharmacological activity with the various extracts,followed by ELISA and fluorogenic quantitative PCR used in studying the mode of components action The results show that:?1?the ethyl acetate was extracted by different polarization from Lepidium latifolium L,five groups???????????were made to get four compounds utilizing chromatographic column and PHPLC from group ?.?2?the consequence of the cell proliferation in vitro shows that the dosages of administration of group ? and ? at 50 ?g/mL?1.172 ?g/mL respectively have significant effect on cell proliferation in the comparison between the rat colonic smooth muscle cells and the DMSO treated group?P<0.01?;dosages of administration of group ? and ? at 50 ?g/mL?37.5 ?g/mL respectively have significant effect on cell proliferation in the comparison between the rat small intestinal smooth muscle cells and the DMSO treated group?P<0.01?.?3?the results of colonic smooth muscle cells ranged by microscopy indicate that compared to the DMSO group,the group ??? both have markable differences?P<0.05?,what's more,there is an extremely significant difference between DMSO group and group ??P<0.01?and a notable difference between DMSO group and group ??P<0.05??4?the ELISA results show that the expression of NO?IP3?Ca2+ in cells ofcolonic smooth muscle and small intestinal smooth muscle treated with the extract and DMSO has no difference?P>0.05?.?5?we get that from the result of qRT-PCR used for gene of ?2 subnit being part of voltage dependence Na+ channel,the expression in colonic smooth muscle cells from group ? and ? has been greater than that in DMSO group?P<0.01?,and the expression in small intestinal smooth muscle cells treated with little and moderate extract in group? is significantly greater than that in DMSO group?P<0.01?and in group ?.The expression in seperated colonic tissues from group of aqueous extract and group ? is greater than that from the control group?P<0.01?,the expression in seperated small intestinal tissues from group of aqueous extract is greater than that from the control group?P<0.01?;the expression in colonic tissues after intragastric administration from group of aqueous extract is greater than that from the control group?P<0.01?;the expression in small intestinal tissues after intragastric administration from group of aqueous extract has no difference with that from the control group?P>0.05?.In conclusion,the effective part of Lepidium latifolium L in group ? helps improve the gastrointestinal motility,and the ?2 subnit being part of the voltage dependence Na+ channel canbe the one of the modes of pharmacological mechanisms of which the targets locate in colonic tissues.
