| Objective To study the improvement of dry eye symptoms in dry eye mice model after giving Yiyanming Oral Liquid as treatment,and the impact on the mRNA and protein expression of some inflammatory factors,including IL-1?,IL 4,TNF-a,in lacrimal gland and cornea of dry eye mice.Methods 30 healthy C57/BL6 mice were divided into 5 groups,which were 1 healthy group and 4 model groups(including 1 control group given oral saline,1 high dose group(320 ? 1/kg),1 middle dose group(160 ? 1/kg)and 1 low dose group(80 ?1/kg)).The healthy group was not given any treatment in the whole 14 days.The other mice in the other 4 groups were given a subcutaneous injection of 0.5 ml of scopolamine hydrobromide at a concentration of 1 mg/ml in the alternating lower limbs 4 times a day(9 o' clock,12 o' clock,13 o' clock,16 o'clock)for 14 days.The amount of tear secretion and corneal fluorescein staining are used as the basis for evaluating the indicators of dry eye symptoms.The control group was given normal saline for 14 weeks.In the test groups,different doses of Yiyanming oral liquid(80 ? 1,160 ?1/kg,and 320 ? 1/kg)were administered to the mice according to their weight.Yiyanman oral liquid was given to different test groups twice per day for 2 weeks.Comparison of clinical effectiveness would be given between the groups during and after the experiment.Results 1.The normal group tear secretion test was 6.04±0.07 mm/min,and the comparison group,low dose group,middle dose group and high dose group were 3.90±0.09mm/min,3.73±0.06mm/min,3.80±0.05mm/min and 3.86±0.02 mm/min.The difference between the normal group and the comparison group,low-dose group,medium-dose group and high-dose group,the difference were statistically significant(P<0.05).There was no statistically significant difference between the comparison group,the low-dose group,the middle-dose group and the high-dose group(P>0.05).It proved the dry eye mice modeled successfully by using scopolamine. 2.After 14 days of treatment in each test group,the observation indexes(tears secretion test,corneal fluorescein staining score)were compared with the comparison group,and the differences were statistically significant.It is proved that Yiyanming Oral Liquid can promote the secretion of tears in dry eye mouse model and reduce the symptoms of dry eye,indicating that Yiyanming oral liquid has therapeutic effect on dry eye mouse model,thus proving that Yiyanming oral liquid has a certain therapeutic effect on dry eye syndrome-In the expression of lacrimal gland and corneal inflammatory factors as IL-4,IL-1 ? and TNF-?,the mRNA expression of high dose group was smaller than that of the control group,and the difference was statistically significant(P<0.05).It indicated that high-dose Yiyanming oral liquid could inhibit the mRNA expression of IL-4,IL-1 ? and TNF-a.The mRNA expression of IL-4 in each experimental group was lower than that in the control group,and the difference was statistically significant(P<0.05).It may be indicated that different doses of Yiyanming oral liquid can inhibit the mRNA expression of inflammatory factor IL-4.4.From the protein expression level of inflammatory factors IL-113 and TNF-?.The protein expression of each experimental group was smaller than that of the control group,and the difference was statistically significant(P<0.05).It is indicated that each dose of Yiyanming oral liquid can inhibit the protein expression of IL-1 ? and TNF-?.The average protein expression of IL-4 in each experimental group was smaller than that in the control group,but the difference was not statistically significant(P>0.05).It indicated that high-dose Yiyanming oral liquid could inhibit the expression of IL-1? and TNF-? in the dry eye mouse model.But the effect of protein expression in IL-4 is not obvious.3.PAS staining showed that the number of goblet cells in the conjunctival descending of the dry eye mouse model of the control group and each experimental group was significantly less than that of the normal group.The number of goblet cells in the descending conjunctiva of the three experimental groups was compared with each other,and the high dose group>middle dose group>low dose group>control group was observed.Conclusion Yiyanming Oral Liquid has a certain therapeutic effect on dry eye syndrome on mice model,by changing the expression of interleukin protein in lacrimal gland and cornea of dry eye syndrome mice model,thus proves that Yiyanming Oral Liquid decreases inflammation on dry eye syndrome. |
