Salvia miltiorrhiza is one of the earliest Chinese herbal medicines recorded in Shen Nong's Herbal Classic.It is widely used for the prevention and treatment of cardiovascular and cerebrovascular diseases.Purpose of this study is to investigate the effects of Salvia miltiorrhiza injection on the proliferation and differentiation of neural stem cells derived from cerebral cortex of fetal mice,as well as its relevant mechanisms.The study will provides some effective experimental data for Salvia miltiorrhiza serving as a potential drug for neurogenesis in the future.This study mainly includes the following aspects:1.Establish a platform for isolation,culture,differentiation and identification of embryonic mouse cerebral cortical neural stem cells to provide a stable cell model for the following studies.2.Study on the effects of Salvia miltiorrhiza injection on the proliferation and differentiation of NSCs: NSCs were divided into two groups,control and Salvia miltiorrhiza injection treatment group.CCK-8 and BrdU Kits,CQ1 Confocal Quantitative Image Cytometer and RT-PCR were used to investigate the drug's effect on NSCs' proliferation and differentiation.Results showed that Salvia miltiorrhiza injection can not only improve the proliferation of NSCs,but also its differentiation into neurons.3.Explore the protective mechanism of Salvia miltiorrhiza injection on NSCs:NSCs were divided into three groups,control,lipopolysaccharide(LPS)treated group and(LPS plus Salvia miltiorrhiza injection)treated group.The result of transcriptome sequencing showed that Salvia miltiorrhiza injection can protect the injured neural stem cells through glutathione metabolism and Ferroptosis,which may be associated with scavenging free radicals and blocking the apoptotic signaling pathway.From the inhibitor experimental result,it can be known that glutathione transferase inhibitor can totally inhibit the protective effect of Salvia miltiorrhiza injection on LPS injured NSCs.Also,it verified the conclusion that the protective mechanism of Salvia miltiorrhiza injection on NSCs is closely related to glutathione metabolism.
