Experimental Study On The Effects Of Bushen Tongqiao Recipe On The Expression Of ERK1/2 In Cochlea Tissue Of The Kidney Ying Deficiency Rats

Objective:ERK1/2,a key factor in MAPK signaling pathway,was used as the core index to study the function and morphological changes of kidney and cochlea in rats after kidney yin deficiency and kidney tonifying prescription intervention,and to explore the relationship between kidney and ear.Part of the molecular mechanism of the relationship between them is explained from the microbiological point of view,which provides experimental basis for modern interpretation of the theory of kidney-dominated ears in Chinese medicine,and provides a new way of thinking for the treatment of inner ear diseases by integrating traditional Chinese medicine and western medicine.At the same time,enrich the theory of Tibetan orifice of traditional Chinese medicine,and provide modern scientific basis for the theoretical development of Tibetan image theory.Methods:SD rats?60 rats?with sensitive auricle reflex and normal ABR threshold were divided into 3 groups?blank group,model group and traditional Chinese medicine group with 20 rats each?according to random numerical table method.The rat model of kidney yin deficiency was established by intramuscular injection of hydrocortisone acetate(25 mg·kg^-1·d^-1)for 7 days in the traditional Chinese medicine group and the model group.Normal saline(5 mL·kg^-1·d^-1)was injected into the blank group.After the successful establishment of the model,the Chinese medicine group was given Bushen Tongqiao recipe(10 mL·kg^-1·d^-1)for 7 days,and the blank group and the model group were given the same amount of normal saline.Observe and remember the general situation of the three groups of rats was recorded.After administration,the auditory function of each group was detected by ABR method,the contents of cAMP and cGMP in plasma were measured,the morphological changes of kidney and cochlea were observed under light microscope,and the expression of ERK1/2 mRNA in cochlea tissue of rats was detected by RT-PCR method.Results:?1?On the index evaluation of kidney yin deficiency in rats,compared with the blank group,the rats in the model group and the traditional Chinese medicine group showed a series of symptoms,such as restlessness,easy to panic,stool dry knot,decrease in eating,drinking water,urine loss,weight loss,and so on.The value of plasma cAMP/cGMP increased?P<0.01?,suggesting that the model of kidney-yin deficiency was successful.However,compared with the model group,the general state of the rats in the traditional Chinese medicine group was better and the plasma cAMP/cGMP value was significantly lower than that in the model group?P<0.01?.?2?Compared with the model group,the ABR threshold in the traditional Chinese medicine group was significantly higher than that in the blank group?P<0.01?.The ABR threshold of the middle model group was the highest.?3?Compared with the blank group,the kidney and cochlea tissue morphology of the model group and the traditional Chinese medicine group showed pathological changes such as glomeruli,renal interstitial fibrosis,renal tubule dilatation,steatosis and so on.The number of hair cells decreased irregular arrangement and the number of spiral ganglia decreased in cochlear tissue.However,the morphology of kidney and cochlea in the traditional Chinese medicine group was better than that in the model group as a whole.?4?The expression of ERK1/2 in cochlea tissue:compared with the blank group,the expression of ERK1/2 mRNA in the model group and the traditional Chinese medicine group was lower than that in the blank group?P<0.05?,but the expression of ERK1/2mRNA in the traditional Chinese medicine group was relatively higher than that in the model group?P<0.05?.Conclusion:After deficiency of kidney yin,it can lead to pathological changes of kidney and cochlea and hearing dysfunction.Bushen Tongqiao recipe can significantly improve the hearing function of rats with deficiency of kidney yin.Its molecular biological mechanism may be that the down-regulation of ERK1/2 expression,the key factor in MAPK signal pathway,leads to auditory signal transduction disorder,leads to apoptosis of cochlear tissue cells,and ultimately leads to hearing impairment and even deafness.ERK1/2 signal pathway is one of the microscopic pathways of kidney and ear.According to the pathological basis of yin deficiency and blood stasis in traditional Chinese medicine,Bushen Tongqiao recipe can up-regulate the expression of ERK1/2 signal,inhibit apoptosis and play an effective role in intervention.
