Objective:To observe the effects of Erzhi Tiangui Granules on quality of oocyte in IVF-ET patients with kidney qi deficiency type of infertility and follicular granulosa cells in patients with SCF mediated the relationship between the signal path, from the perspective of inhibition of granular cell apoptosis to discuss kidney in traditional Chinese medicine to improve the mechanism of action of IVF-ET in patients with pregnancy outcome.Methods:Eighty patients undergoing IVF-ET in patients with kidney qi deficiency were selected, according to the number of visiting time, odd number for treatment group (erzhi Tiangui granule group), even numbers as the placebo group, each with 40 cases in each. Select another 40 cases of female roughly normal, due to male factors of IVF-ET patients, as control group. The treatment group began to take Erzhi Tiangui granules from the follicular phase, the placebo group and control group at the same time taking a placebo. Compare the syndrome integral change of kidney qi deficiency and infertility patients; Gonadotropin (Gn) days and the total amount of drug use; HCG day blood E2, LH, P level; High quality oocyte rate; High quality embryo rate; clinical pregnancy rate. Three groups of granulosa cells were collected on the oocyte retrieval day, and mRNA Bim expression was determined by real-time fluorescence quantitative method (real-time, PCR) in three groups of patients with follicular granulosa cells, meanwhile the expression of p27kip1 Protein was determined by blot Western method.Results:(?) 1. Before and after treatment, the difference of kidney qi deficiency syndrome integral in Erzhi Tiangui granule group is statistically significant (P< 0.05);There was no significant difference in the syndrome of kidney qi deficiency before and after treatment in the placebo group (P> 0.05); The difference between the two groups before and after treatment of kidney qi deficiency syndrome, the difference was statistically significant (P< 0.05).2.Compared with the placebo group, Erzhi Tigui granule group and non kidney deficiency group GN dosage were lower than in the placebo group, the difference is significant (P< 0.05); Erzhi Tigui granule group and non kidney deficiency group difference no significant (P> 0.05).3.There was no significant difference in the serum levels of E2, LH, and P in HCG days between the three groups (P>0.05).4. Compared with the placebo group, the difference of high-quality oocytes rate, good quality embryo rate, clinical pregnancy rate is significant (P< 0.05) that Erzhi Tiangui treatment group was higher than non kidney deficiency group;There was no significant difference between Erzhi Tiangui treatment group and non kidney deficiency group. (?) 1. Compared with the placebo group, BIM mRNA expression and p27kipl Protein expression of Erzhi Tiangui treatment group and non kidney deficiency group were increased,and the difference is significant (P<0.05)Conclusions:Traditional Chinese medicine of Tonifying the kidney to the upregulation of Bim and p27kipl gene expression through the regulation of SCF mediated signal transduction pathway, so as to improve the quality of oocyte, improve kidney yin deficiency in patients with infertility IVF pregnancy outcome.
