Objective:The diabetic(DM)model rats induced by streptozotoxin(STZ)were taken as the object of study,and the changes of serum S-Cr,BUN content,urine ALB content and renal histomorphology of rats were taken as indexes.To investigate the renal protective effect of Mongolian medicine Xierga-4 wei decoction on diabetic rat model.The changes of blood glucose and body weight in rats were observed,and the hypoglycemic effect and effect on body weight of Mengyao Xirga Siwei decoction were discussed,which provided reliable experimental basis for clinical application and further research and development of Mengyao Xirga Siwei decoction.Methods:(1)48 male Wister rats with 200±10 g Wister were randomly divided into 6 groups:normal group,DM model group,positive control group and Mongolian medicine Xierga-4 wei decoction,which were randomly divided into 6 groups:normal group,DM model group,positive control group and Mongolian medicine Xierga-4 wei decoction.Low dose group.In addition to the normal group,the other groups were given a single intraabdominal injection of STZ 50mg·kg-1 on an empty stomach for 12 hours,followed by a normal diet.72 hours after injection of STZ,blood samples were collected from the tail vein of rats.Empty abdominal blood glucose and urine glucose were measured.Fasting blood glucose?16.7 mmol/L,urine sugar above.That is to say,the DM rat model was successfully established.2 the method of administration:the normal group and the model group were given the same amount of 20ml kg 1·d 1 distilled water,the positive control group was given enalapril 1.5mg kg 1·d 1 intragastrically,and the Mongolian medicine Xierga-4 wei decoction was high and medium.In the low dose group,1.8 g·kg-1-d-1,0.9 g·kg-1 d-1,0.45 g·kg-1·d-1 were given intragastrically,and each group was given a continuous dose of 1.8 g kg 1·d 1,0.9 g kg 1·d 1 and 0.45 g kg 1·d 1,respectively.In the course of the experiment,fasting blood glucose and urine glucose were measured regularly.If hypoglycemia occurred,the number of diet should be increased appropriately to avoid adverse effects on the experimental results.(2)observation indexes and determination:1 during the experiment,The basic status and body weight of rats in each group were recorded.2 the changes of blood glucose and urine glucose were measured regularly before and after modeling and after administration.3 after 8 weeks of administration,the rats were randomly collected and centrifuged with 10min(3000 r·min-1,4?)and stored at-80?.The rats were killed,blood samples were collected from abdominal aortic,and serum 10Omin(3000 r·min-1,4?)was isolated by centrifugation.The contents of serum S-Cr,BUN and urine ALB were measured by ELISA.4 the pathological changes of renal tissue were observed.(3)Statistical processing:the results of the measurement data were expressed by X±S,and the statistical analysis was carried out by SPSS17.0 software.The measurement data were analyzed by single factor variance analysis and multiple comparisons,and the differences were statistically significant(p<0.05,p<0.01).Results:(1)The effect of Mengyao Xierga-4 wei decoction on serum S-Cr,BUN content and urine ALB content in DM rats:compared with the model group,the serum S-Cr,BUN content and urine ALB content in each administration group single factor variance analysis and multiple comparisons,and the differences were statistically significant(p<0.05,p<0.01).Results:(1)the effect of Mengyao Xiriga Siwei decoction on serum S Cr,BUN content and urine ALB content in DM rats:compared with the model group,the serum S Cr,BUN content and urine ALB content in each administration group decreased.Among them,the positive control group and Xiezha 4 Wei decoction high dose group decreased significantly,there was significant difference(p<0.05).(2)on blood glucose in DM rats:compared with the model group,4 weeks after administration and 8 weeks after administration,The blood glucose of rats in each administration group decreased,especially in the positive control group and the high dose group of Xieriga 4 Wei decoction.There was significant difference(p<0.05).(3)on the pathological changes of renal tissue in DM rats.In the model group,no obvious pathological changes were found in the glomeruli,and more vacuolated vacuole degeneration and necrosis were found in the renal tubules,and there were significant differences in the pathological changes of the renal tissues in the model group.There were protein tube type and cell tube type in renal tubule cavity.In the western medicine group,some renal tubules showed cavitation vacuole degeneration and necrosis of epithelial cells,while in the low dose group,part of renal tubules showed cavitation vacuole degeneration and necrosis of epithelial cells,which was worse than that of western medicine in part of renal tubules,and in the low dose group,the cavitation vacuole degeneration and necrosis of epithelial cells in some renal tubules were worse than those in western medicine.In the middle dose group of Mengyao Xiriga 4 Wei decoction,more renal tubules showed cavitation vacuolation of epithelial cells,which was worse than that of the low dose group.In the high dose group of Mengyao Xiriga 4 Wei decoction,the cavitation vacuolation of renal tubular epithelial cells was occasionally observed,and the degree of renal injury was the least compared with other groups.Conclusion:Mongolian medicine Xiriga 4 Wei decoction has certain renal protective effect on diabetic rats,and Mongolian medicine Xirga 4 Wei decoction has different degrees of hypoglycemic effect.
