ObjectiveTo explore the new extraction process of Tremella Polysaccharides and obtain the best extraction conditions of the wet broken assisted ultrasonic method;To establish mice model with outer-dry skin that evaluate the skin Comprehensively and to study the effect of intervention with Tremella Polysaccharides;To explore the mechanism of action of Tremella polysaccharides in mice with Outer-dry Skin.Methods1.Compared with the traditional extraction method,the wet broken assisted ultrasonic method demonstrates the high efficiency of the new process by measuring the extraction yield,polysaccharide content and total sugar yield and orthogonal experiment was further used to determine the optimal extraction conditions of the new process;2.The mice model with Outer-dry Skin was established according to the comprehensive conditions of "temperature-humidity-wind" in the artificial climate chamber(temperature 22±2?,relative humidity 33±2%,temperature and humidity remain unchanged,wind speed 2.5 m/s,wind treatment time 5 h/d),and then evaluated the dryness skin Comprehensively;to observe the effects of apparent characteristics,water content and tissue structure of mice by oral and topical Tremella polysaccharides;3.The effects of Tremella polysaccharides on skin and lung tissue structure,HA content and AQP3 expression in mice were observed respectively by HE staining,ELISA and immunohistochemistry.Results1.Compared with other methods,the Wet Broken Assisted Ultrasonic Method will significantly increase the extraction yield and polysaccharide content of Tremella(p<0.01),and increase the total sugar yield significantly(p<0.05),which has statistical significance.The optimum extraction conditions of the Wet Broken Assisted Ultrasonic Method were as follows:broken wall time 8 min,temperature 50 0C,liquid material ratio of 50:1(g/mL)and ultrasonic time 15 min.The order of influence of each factor is:liquid to material ratio>wall breaking time>ultrasonic time>extraction temperature;2.After comprehensive evaluation of skin,the external dryness model was established successfully.Under the intervention of Tremella polysaccharides,compared with the model group,the water consumption of the mice in the oral and topical administration groups decreased,and the degree of dryness of the stool was relieved,the skin of each drug group became sleeker and glossier,without obvious dandruff and crack,the hair of mice grew normally.Under light microscope,the skin tissue structure of each group was relatively complete compared with the model group,the stratum corneum decreased exfoliation,and the number of hair follicles and sebaceous glands increased.Compared with the blank group,the skin water content of the model group with oral and topical administration was decreased(P<0.05).Compared with the model group,the skin dryness of each drug group was relieved(P<0.05),and the effect of 400 mg/kg TP group was the best among oral groups and the topical TP group was better than that of the positive control group among the external administration groups(P<0.05);3.After modeling,compared with the blank group,,the mice in the model group had easier to fall off and wrinkled stratum corneum of the skin,less hair follicles and sebaceous glands and more obvious proliferation of connective tissue under the microscope;the interval between the lungs was widened and congestion occurred,and some severe cases showed pulmonary edema.After the intervention of Tremella polysaccharides,compared with the model group,the skin and lung tissues of the mice in the oral and topical administration groups were more complete under the microscope,and the HA content in the skin and lungs was significantly up-regulated(P<0.05).Except for the low-dose group of Tremella polysaccharides,the expression of AQP3 in the lungs was not significantly different from the model group(P>0.05),but the AQP3 in the skin and lungs of the other groups was significantly increased(P<0.05).Conclusion1.As a new process,the wet broken assisted ultrasonic method that has the advantages of simple operation,low cost,high extraction rate and short time consum--ing is not only a composite method but also a technical innovation compared with other traditional processes.It provides a reference for further research and development of Tremella polysaccharides;2.The successful modeling of the mice with outer-dry skin suggests that the theory of skin dryness based on external dryness is established,and Tremella polysaccharide can alleviate dryness of skin effectively and has certain curative effect on model mice with outer-dry skin;3.Based on the theory of lung main fur",experiments had shown that Tremella fuciformis can improve the pathological structure of skin and lung tissue of mice with skin dryness,and simultaneously up-regulate the expression levels of HA and AQP3 in skin and lung.There may be a close relationship between skin and lungs of the model mice.Tremella polysaccharides had a regulating effect on the lung and skin of the model mice,and its mechanism of action may be related to HA and AQP3.
