Backgroud:Endometriosis(EMs)is a common gynecological disease among women of child-bearing period.Although the lesion is benign,it has a serious impact on the physical and mental health of women of child-bearing period.In recent years,the incidence of endometriosis has increased,and the malignant transformation of endometriosis deserves special attention,Endometriosis occurs mostly in the ovary.Malignant transformation of endometriosis of ovary also called endometriosis associate ovarian cancer(EAOC).Although the prognosis of patients with EAOC is better than that of other types of ovarian cancer,the early onset of the disease has a great influnce on the quality of life of patients.Early treatment supervision and early diagnosis of EAOC will benefit EAOC patients.Lin28 is an RNA-binding protein,which has become a new hotspot in cancer research.The expression of Lin28 in ovarian cancer and other malignant tumors is closely related to the prognosis of cancer patients.More importantly,Lin28 is involved in Let-7 carcinogenic pathway,which is well known to us.HMGA2 protein is a DNA binding protein,which interacts with many carcinogenic genes to participate in the process of invasion,metastasis,malignant tumors progression and drug resistance.The expression of Lin28 and HMGA2 proteins in EAOC will provide an important basis for further study of Lin28 and HMGA2 proteins in endometriosis associate ovarian cancer Objective:The main objective is to investigate the expression of Lin28 and HMGA2 proteins in EAOC,and to analyze the relationship between target proteins and EAOC.we will collect clinical dates to investigate the correlation between the expression of the two proteins in EAOC and athological stage,differentiation,prognosis,which may provide the possibility for early diagnosis and prognosis of EAOC through pathogenesis of proteins.In addition,this study will analyze the correlation betweenthe two proteins expressed in EAOC and provide a basis for the study of the malignant transformation mechanism in EAOC.Methods:Twenty patients with EAOC from January 2012 to December 2017 in the Jilin University Second Hospitalwere selected as the experimental group,and 20 patients with ovarian endometriosis cyst and normal endometrium were selected as the control group.Pathology of patients in case group and control group was stained by immunohistochemical method.The staining results are analyzed by Motic Images Advanced 3.2 image analysis software,result of experiment is reprensent by the optical density value.The measured optical density values can represent the expression levels of Lin28 and HMGA2 proteins in ovarian cancer and endometrium tissues of EAOC patients,ovarian endometriosis cyst and normal endometrium of control group.Results:1.In the group of endometriosis-related ovarian cancer and ovarian endometriosis cyst,there were differences in age,symptoms,tumor size and CA125 value between the two groups,but there were no differences in tumor side,menstruation,secondary dysmenorrhea,pregnancy and abortion history between the two groups.2.The expression levels of Lin28 and HMGA2 protein in EAOC were significantly higher than those in ovarian endometriosis cyst,and the difference was statistically significant.3.The expression of Lin28 and HMGA2 protein in endometrium of EAOC patients was significantly higher than that of normal endometrium group,and the difference was statistically significant.4.The expression of Lin28 and HMGA2 in malignant ovarian and corresponding endometrial tissues of EAOC has a linear positive relationship.5.The expression of Lin28 and HMGA2 in ovarian endometriotic cysts was higher than that in normal endometrial tissues,and the difference was statistically significant.6.The expression levels of Lin28 and HMGA2 were not correlated with EAOCtumor size,preoperative CA125 value,patient age and EMs history,but with FIGO stage,tissue differentiation and recurrence.Conclusion:1.Theage,CA125 valueandtumor diameterof patients with endometriosisrelated ovarian cancer were higher than those of patients with ovarian endometriosis cyst,and the clinical symptoms were more common.2.The expression of Lin28 and HMGA2 proteins in ovarian cancer tissue of EAOC is significantly higher than that in ovarian endometriosis cyst group,and in endometrial tissue of EAOC patients is significantly higher than that of normal endometrium control group.Therefore,Lin28 and HMGA2 proteins are related to the occurrence and development of EAOC.3.Lin28 and HMGA2 proteins were correlated with the occurrence and development of EAOC.Lin28 and HMGA2 proteins may be involved in the occurrence of endometriosis.4.In EAOC patients,the later the FIGO staging,the worse the differentiation of cancer tissue,the higher the expression of Lin28 and HMGA2 protein,which mean the greater the possibility of recurrence.
