Sweet tea(Rubus suavissimus S.Lee)was included in the“Guangxi Zhuang Medicine Quality Standard(Volume 2)”.Sweet tea is a Chinese herbal medicine resource that combines the identity of sugar,tea and medicine.It is sweet,and it belongs to the liver,lungs and bladder.It has the pharmacological effects of clearing away heat and nourishing both lungs,removing cough and relieving cough,regulating airways and waterways.Pharmacological effects of sweet tea showed that sweet tea had many pharmacological effects such as treating diabetes,regulating blood lipids,anti-allergic,anti-oxidation,protecting liver tissue,strengthening immunity,and anti-inflammatory and antibacterial.1.In this experiment,sweet tea was used as a raw material,sweet tea was pulverized,extract with 40%ethanol,and the extract of sweet tea was concentrated to obtain an extract.The sweet tea extract was extracted with different polar solvents,and the extract was separated by silica gel column chromatography and preparative thin layer chromatography.The results of nuclear magnetic spectroscopy analysis showed that the four compounds wereβ-sitosterol(A),rubusoside(B),quercetin(C),Isoquercitrin(D),gallic acid(E)and dibutyl phthalate(F),wherein dibutyl phthalate(F)is first isolated from sweet tea.2.In the experiment,the content of sweet tea was used as the index,and on the basis of single factor,the extraction process of sweet tea was explored by ultrasonic assisted extraction and response surface methodology.The optimal result was that the ethanol concentration was 35.93%,the ratio of material to liquid was 1:29.41,the ultrasonic temperature was 61.64℃,and the ultrasonic time was 30.68 min.And the predicted result was 3.88%.In order to improve the operability of the experiment,he experimental conditions were modified as follow:ethanol concentration was 36%,the ratio of material to liquid was 1:30,ultrasonic time was 62℃,ultrasonic time was 30 min,and the average experimental result of the modified experimental conditions was 3.65%.The percentage error between the actual measured value and the experimental predicted value after modifying the experimental condition was 0.23%.The extraction effect was better than the single factor extraction,the deviation of the fitting result was low,and the experimental results had high reliability.3.In other to improve the effective utilization rate of sweet tea,sweet tea was prepared into granules,and the granule preparation process was investigated from the aspects of molding rate,melting property,fluidity and percentage of moisture absorption.The ratio of sweet tea dry powder to soluble starch was 1:2,and the prepared sweet tea granules have better effects.The experiment examined 3 batches of homemade sweet tea granules,the moisture content was less than 8%,the sum of the coarse particles which can not pass through the No.1 sieve and fine powder that can pass through the No.5 sieve were less than or equal to 15%,and the dissolution test should be dissolved with 5min completely,the granule loading difference was plus or minus 5%,and the granule inspection results were in compliance with the pharmacopoeia inspection specifications.4.The experiment initially explored the protective effect of homemade sweet tea granules on the liver of mice.From the experimental results,the serum biochemical indicators ALT and AST,liver biochemical indicators SOD and MDA,compared with the model group,these biochemical indicators showed the sweet tea granules in the middle dose group and the high dose group were all significant.The experimental results showed taht sweet tea granules had protective and preventive effects on acute liver injury of mouse CCl4.The results of HE staining pathological section of mouse liver tissue also proved that sweet tea granules had preventive and protective effects on acute liver injury in mice.In the above experiment,systematically extracting the chemical components of the sweet tea,exploring the optimal extraction process of the unique components of the sweet tea,preparing and inspecting the homemade sweet tea granules,and preliminary exploring the pharmacological effects of the granules.Through the rational development and utilization of sweet tea resources,it provides a scientific basis for further research on sweet tea granules.
