| Objective:The prevention and treatment of chronic kidney disease(CKD)has become one of the most important public health problems in the world.With the advent of aging in China,chronic renal failure(CRF)has not only become one of the important diseases threatening people's health,but also caused a huge amount of medical expenditure.The main causes include chronic nephritis,diabetes mellitus,hypertensive kidney damage and so on.The disease is concealed in the early morning.Due to the neglect of the individual's physical condition and the neglect of physical examination and condition,most patients' kidney function has been seriously damaged at the first visit.After entering the end stage renal disease(ESRD),western medicine mainly uses alternative therapy to treat chronic renal failure,which is expensive and difficult to popularize.Therefore,traditional Chinese medicine has unique advantages in treating chronic renal failure.Rhubarb has been widely used as an essential drug in the treatment of chronic renal failure,and its effectiveness has been fully confirmed.But throughout the use of Rhubarb in the treatment of chronic renal failure,the masters did not specify the specific use of rhubarb,and how long rhubarb should be decocted is the best.Based on this,this paper intends to explore the effects of Different Decocting Time of rhubarb processed products on diarrhea,intestinal propulsion and Na+-K+-ATPase activity in mice,so as to provide theoretical basis for clinical application of rhubarb.At the same time,the clinical effect of "Tongfu Xiezhuo" method used by Professor Ba Yuanming to delay the progress of CRF was observed.Methods:Four kinds of raw rhubarb,cooked rhubarb,liquor rhubarb and rhubarb charcoal provided by traditional Chinese medicine in Hubei Province were taken.Each portion was added with appropriate amount of water and decocted with direct fire for 5,7,9,10,15,20,30minutes(after boiling,decocting with Wenhuo,calculating the time from the time of medication),and the medicinal liquid was filtered out.The corresponding number of mice were divided into blank group and experimental group with corresponding reagents.The diarrhea and intestinal propulsion of mice were observed,and the data were recorded for statistical analysis.After the experiment,the colon of mice was taken and the activity of Na+-K+-ATPase was determined.To screen 60 cases of "Tongfu Xiezhuo" method adopted by Professor Ba Yuanming in outpatient clinic,and analyze the changes of TCM clinical symptom score,Scr,BUN,UA,e GFR and 24 upq before and after treatment.Result:Rhubarb had the strongest diarrhea effect at each time point.Rhubarb had the strongest diarrhea effect when decocted for 9minutes.EI was 12.8.The strongest intestinal propulsion effect was raw rhubarb,which had the strongest intestinal propulsion effect when decocted for 9 minutes.The intestinal propulsion rate was 84.09%.The lowest activity of Na+-K+-ATPase in the whole is the ripe rhubarb.Na+-K+-ATPase activity of cooked rhubarb was the lowest when it was decocted for 7 minutes,which was 5.After the treatment of "Tongfu Xiezhuo" method,the scores of TCM clinical symptoms,Scr,BUN,UA,24 upq were significantly reduced,and e GFR was also significantly increased.Conclusion:Rhubarb processed by four methods can promote diarrhea(the diarrhea effect is: raw rhubarb > wine rhubarb cooked rhubarb >rhubarb charcoal in turn).Except rhubarb charcoal,three processed products of rhubarb can promote intestinal propulsion(the order of strength is raw rhubarb > cooked rhubarb > wine rhubarb > rhubarb charcoal).The four Processed Rhubarb could inhibit the activity of Na+-K+-ATPase in colon of mice(the degree of inhibition was as follows: cooked rhubarb > raw rhubarb > wine rhubarb > rhubarb charcoal).The diarrhea effect of rhubarb changed with the prolongation of decocting time.By observing the clinical effect of "Tongfu Xiezhuo" method,it is proved that it has better effect on relieving clinical symptoms and delaying the progress of renal function. |
