Objective:The purpose of this experiment was to evaluate the therapeutic effect of the traditional Chinese medicine?Bushen-conger Recipe which is used for treating sensorineural deafness?on the drug-induced deafness,and to study the effect of the prescription on the apoptosis signal pathway P38MAPK.By comparing the general state of the three groups of rats,changes in body weight,hearing,blood cAMP and cGMP levels,cochlear pathological sections and the exprssion of cochlear P38 protein,to assess the overall therapeutic effect of Bushen-conger Recipe.Provide a laboratory basis for the clinic to use Bushen-Conger method to prevent and treat drug-induced deafness.Methods:After screened by ABR,forty-five SD rats were divided into three groups?15rats in each groups?by the random number table?control group,GM model group and TCM group?.The rat model of drug-induced deafness was prepared by given intramuscular injection of gentamicin(100 mg·kg-1·d-1)for 14 consecutive days.The Chinese medicine group was given intragastric administration of Bushen-conger Recipe on the basis of model building.The blank group was injected with the same amount of normal salt?NS?water.We observed the rat's general conditions,difference in body weight gain,the extent of hearing loss,and the blood cAMP and cGMP contents.The pathological changes of the rats'cochlea were observed by H-E staining and the content of P38 protein in rats'cochlea was detected by Western-Blot and RT-PCR.Results:?1?General status evaluationThe general state of the rats in the model group and the TCM group has changed,but the rats in the blank group performed normally.Both rats of the Traditional Chinese medicine group and the model group have the following symptoms:restlessness,easily frightened,decrease of the food-intake,dried stools and increase of the daily drinking water.Meanwhile,the body weight increased slowly,and the urine volume decreased significantly.Rats in the Chinese medicine group appeared like the model group,but the symptoms were significantly relieved.?2?Weight changesThe body weights of the three groups were evaluated after two weeks.Compared with the model group and the traditional Chinese medicine group,the blank group had the fastest weight gain?P<0.01?.Compared with the traditional Chinese medicine group,the weight gain of the model group was slower?P<0.01?.?3?Comparison of blood cAMP and cGMP valuesThe cAMP/cGMP value of the model group was higher than that of the blank group?P<0.05?.The cAMP/cGMP value of the Chinese medicine group was higher than that of the blank group?P<0.05?.Compared with the Chinese medicine group,the cAMP/cGMP value of the model group was higher in the model group?P<0.05?.?4?Comparison of ABR thresholdCompared with the model group and the Chinese medicine group,the blank group had the lowest ABR threshold?P<0.01?,suggesting that the blank group had the best hearing ability,the model group had a lower ABR threshold value compared with the traditional Chinese medicine group,suggesting that rats'hearing in the Chinese medicine group was better than model group?P<0.01?.?5?Comparison of the cochlea's pathologicalThe blank group rats had normal cochlear sections.Rats in the model group and the Chinese medicine group showed a decrease in the number of hair cells and spiral ganglions,the arrangement of the hair cells is irregular.On the whole,the pathological section of the Chinese medicine group is better.?6?The expression of P38 protein in cochlearResults of Western-Blot and RT-PCR both showed that compared with the model group and the traditional Chinese medicine group,the expression of P38 protein in the blank group was lower?P<0.05?,while the expression of the P38 protein in the Chinese medicine group was relative Lower to that of the model group?P<0.05?.Conclusion:The general states,body weight changes,and cAMP/cGMP values indicated that the overall state of the two groups of rats in the model group and the Chinese medicine group was similar to the syndrome of kidney deficiency.Therefore,the method of tonifying kidney to treat drug-induced deafness was feasible.The ABR thresholds and pathological sections of the three groups of rats showed that the rat model of drug-induced deafness was successful.At the same time,the improvement of the ABR threshold and the results of pathological sections of the cochlea suggest that Bushen-conger Recipe has a certain degree of prevention and control of drug-induced deafness in rats.The expression of P38 protein in the cochlea of three groups of rats suggested that the pathogenesis of drug-induced deafness induced by gentamicin may be related to the initiation of apoptosis in MAPK cells.The use of Bushen-conger Recipe can effectively inhibit the opening of the P38MAPK channel and thus play a significant role on drug-induced deafness.
