Objective:The aim of this study was to observe the effects of acupuncture preconditioning on acute myocardial ischemia-reperfusion injury(myocardial ischemia-reperfusion injury,MIRI)in rats with acute myocardial ischemia-reperfusion injury(Electrocardiogram ECG),cerebellar nucleus and lateral hypothalamic area,Dopamine,DA),5-hydroxytryptamine(serotonin),5-hydroxytryptamine(5-hydroxytryptamine,serotonin 5-HT)neurotransmitter content and the effect of c-fos protein on the cerebellar parietal nucleus-lateral hypothalamus The role and mechanism of acupuncture in anti-acute MIRI effect and the possible material basis of cerebellar-hypothalamus involvement in anti-myocardial ischemia-reperfusion injury were preliminarily investigated.Methods: 72 healthy male SD rats were divided into "sham operation group","model group","electro-acupuncture heart meridian group"(referred to as "heart meridian group"),and 12 rats in each group in electro-acupuncture lung meridian group(referred to as "lung meridian group").They were divided into two groups: "lesion of the lateral hypothalamic area and electroacupuncture of the cardiac meridian"("LHA's meridian")and lesion of the cerebellar parietal nucleus("FN's meridian"),with 6 rats in each group.The acute MIRI rat model was established by ligation of left anterior descending coronary artery in the model group without electroacupuncture."Heart Classic Group" selected acupoints "LT7" points,"LT5" points,"Lung Classic Group" selected acupoints "LU9 " Acupoint," LU7 " point,stimulated current 2m A,the frequency of 2 Hz,each 15 minutes,continuous electro-acupuncture intervention for 7 days." LHA heart meridian " group,rats after brain localization,microinjection of(kainic acid,KA)lg/L solution 0.4 uls.Observe 3 days later,electroacupuncture intervention 7 days."FN heart meridian" group,damage electroacupuncture method and "LHA heart meridian" group.The acute MIRI model was made within 1 hour after the last electroacupuncture,and the changes of ECG were recorded at the same time.After the successful replication of the model,microdialysis technique was used to study the intracellular fluid in the brain of each group of rats.The content of DA,5-HT neurotransmitter in nucleus was determined by high performance liquid chromatography-electrochemical detection system.The contents of c-fos protein in cerebellar parietal nucleus and lateral hypothalamic area of each group were detected by immunohistochemical method.Spss22.0 software was used for statistical analysis and graphic production.Results: 1.Comparison of ST Segment displacement of ECG in different groups of Rats:There was no significant difference in the displacement of ST segment between the three groups before replication(P > 0.05).Except for "sham operation group",there was significant difference in the displacement of 30 min ST segments at 10 min,after ligation compared with that before model replication(P < 0.05),and there was no significant difference between the two groups(P < 0.05).Except for "sham operation group",the displacement of ST segment at 30 min after reperfusion was significantly higher than that at 10 min after ligation(P < 0.05).At 30 min reperfusion,compared with "model group","heart meridian group","LHA meridian group"," The displacement of ST segment in the FN heart meridian group was significantly lower than that in the heart meridian group(P < 0.05),and compared with the "heart meridian group",the ST segment displacement in the LHA heart meridian group was significantly higher than that in the "heart meridian group"(P < 0.05).2 Comparison of the contents of DA and 5-HT in cerebellar parietal nucleus of rats in each group 2.1 Comparison of DA content in cerebellar parietal nucleus of rats in each group: Compared with the sham operation group,the content of DA in the cerebellar parietal nucleus in the model group was significantly lower than that in the sham operation group(P < 0.05).Compared with the "model group",the content of "heart meridian group" was significantly higher than that of the "model group"(P < 0.05).Compared with sham operation group,the content of DA in cerebellar parietal nucleus in lung meridian group and LHA heart meridian group was significantly lower than that in heart meridian group(P < 0.05).2.2 Comparison of 5-HT content in cerebellar parietal nucleus of rats in each group: Compared with the sham operation group,the content of 5-HT in the cerebellar parietal nucleus in the model group was significantly lower than that in the sham operation group(P < 0.05).Compared with the "model group",the content of 5-HT in the lateral hypothalamic area in the "Xinjing group" was significantly higher than that in the "model group"(P < 0.05).Compared with the "heart meridian group",the content of 5-HT in the lateral hypothalamic area in the "lung meridian" group and "LHN heart meridian" group was lower than that in the "heart meridian group"(P < 0.05).3 Comparison of the contents of DA and 5-HT in the lateral hypothalamic area of each group of rats 3.1 Comparison of DA content in lateral hypothalamic area of rats in each group: Compared with the sham operation group,the content of DA in the lateral hypothalamic area in the model group was significantly lower than that in the sham operation group(P < 0.05).Compared with the "model group",the content of DA in the lateral hypothalamic area in the "heart meridian group" was significantly higher than that in the "model group"(P < 0.05).Compared with the "heart meridian group",the content of DA in the lateral hypothalamic area in the "lung meridian" group and "FN heart meridian" group was higher than that in the "heart meridian group",and there was no statistical significance(P > 0.05).3.2 Comparison of 5-HT content in lateral hypothalamic area of rats in each group:Compared with the sham operation group,the content of 5-HT in the lateral hypothalamic area in the model group was significantly lower than that in the sham operation group(P < 0.05).Compared with the "model group",the content of 5-HT in the lateral hypothalamic area in the "Xinjing group" was significantly higher than that in the "model group"(P < 0.05).Compared with the "heart meridian group",the content of 5-HT in the lateral hypothalamic area of the lung meridian group and the FN heart meridian group was lower than that of the "heart meridian group"(P < 0.05).4 Expression of c-fos protein in cerebellar parietal nucleus and lateral hypothalamic area of rats in each group 4.1 Expression of c-fos protein in cerebellar parietal nucleus: Compared with the sham operation group,the positive expression of c-fos protein in the cerebellar parietal nucleus in the model group was significantly higher than that in the model group(P < 0.05),and the positive expression of c-fos protein in the cerebellar parietal nucleus in the "heart meridian group" was significantly lower than that in the "model group"(P < 0.05).Compared with the "heart meridian group",the positive expression of c-fos protein in the cerebellar parietal nucleus in the lung meridian group and the LHA heart meridian group was significantly higher than that in the "heart meridian group"(P < 0.05).4.2 Expression of c-fos protein in lateral hypothalamic area:Compared with the sham operation group,the positive expression of c-fos protein in the lateral hypothalamic area in the model group was significantly higher than that in the sham operation group(P < 0.05).Compared with the "model group",the positive expression of c-fos protein in the lateral hypothalamic area of the "heart meridian" group and "FN heart meridian" group was significantly decreased(P < 0.05).Compared with the "heart meridian group",the positive expression of c-fos protein in the lateral hypothalamic area of the "lung meridian" group and "FN heart meridian" group was significantly higher than that of the "heart meridian group"(P < 0.05).Conclusion: 1.The fastigial nucleus-lateral hypothalamic area participates in the mechanism of acupuncture of the heart meridian to improve the acute MIRI effect.2.In the fastigial nucleus of cerebellum and lateral hypothalamus,monoamine neurotransmitters DA and 5-HT may be the important material basis for acupuncture to improve acute MIRI.
