Objective:To study the effect of Matrine on TGF-?/Smad2 signaling pathway in rat pancreatic fibrosis and explore the role of matrine in pancreatic fibrosis.Methods:1?Establishment of a rat model of pancreatic fibrosis;2?According to the purpose of the study,the experiment was divided into three groups: sham operation group,model group and model + matrine group;3?HE was used to detect the pathological results of each group and to judge whether the model was successful or not;4 ? Immunohistochemistry was used to detect the expression of ?-SMA,TGF-?R1 and TGF-?R2 collagen in pancreatic tissue of each group;5?The expression of Smad2,T?R? and T?R? in pancreatic tissues of each group was detected by fluorescence quantitative PCR and Western blot.Result:1?After the model construction,One rat died in the model group and one rat in the Matrine group.The results of HE showed the hyperplasia of small glands around the gland,the swelling of the acinar cell mitochondria,the accumulation of a small amount of interstitial fibers and no cell infiltration.The above results show that the model is constructed successfully.In sham operation group,there was no obvious fibrosis and no progressive changes.In matrine group,a small amount of acinar cells mitochondria swelled and the basic structure was completed.2?Compared with sham operation group,the expression of ?-SMA,TGF-?1 and I collagen in model group increased significantly,and the difference was statistically significant(P < 0.05).3?Compared with the model group,the expression of ?-SMA,TGF-?1 and I collagen in the Matrine group decreased significantly,and the difference was statistically significant(P < 0.05).4?Compared with sham operation group,the expression of Smad2,T?R?,T?R? collagen in model group increased significantly,and the difference was statistically significant(P < 0.05).5?Compared with the model group,the expression of Smad2,T?R? and T?R? in matrine group decreased significantly,the difference was statistically significant(P < 0.05).Conclusion:The TGF-/Smad2 signaling pathway may be involved in the process of pancreatic fibrosis.Matrine can inhibit the activation of pancreatic stellate cells by down-regulating the expression of TGF-beta 1 and Smad2,which may inhibit the occurrence and development of fibrosis through this mechanism,providing a theoretical basis for matrine's clinical application..
