Objective:To observe the curative effect of Jiajiansanjiasan on rats with hepatic fibrosis,and uncover the molecular mechanism of action based on analysis of inflammatory factors and TGF-β1/Smad2 signaling pathway, finally to explore the modern biological basis of Zhukejiao pathological theory.Methods:24 Wistar rats were randomly divided into control groups, model groups, positive control groups and Jiajiansanjiasan groups. After 7days common breeding,all the rats were duplicated immuno-hepatic fibrosis model by injecting porcine serum intraperitoneally except the control groups.After modeling,all the rats were taken medicine by gavage, once daily.The dosage of Jiajiansanjiasan groups was 8.125g·kg-1, positive control groups was 0.625g·kg-1,control groups and model groups were taken with sovolumetric 0.9%NaCl.After 60days remedy, taking rats liver, dyeing by HE and MASSON, testing hepatic fibrosis indexes, IL-1, IL-6, TGF-a by ELISA method, detecting TGF-β1/Smad2 by RT-PCR method.Results:1. The hepatic tissue of the control group was normal, the hepatic lobule structure were integrated, without liver cell degeneration or necrosis, without fibrous connective tissue hyperplasia. The hepatic tissue of the model group appeared cellular swelling, ballooning degeneration, lipoid degeneratiaon, large scale of fibrous tissue proliferation from central veins to portal area, formation of pseudolobuli. Jiajiansanjiasan group and positive control group also appeared cellular swelling, small scale of fibrous tissue proliferation around portal area. Compared with control group, the liver tissue inflammation activity grading, hepatic fibrosis staging and collagen fiber area percentage increased significantly in model group and Jiajiansanjiasan group and positive control group rats. Compared with model group, the liver tissue inflammation activity grading, hepatic fibrosis staging and collagen fiber area percentage decreased significantly in Jiajiansanjiasan group and positive control group rats.2. Compared with control group, serum HA, LN, PC Ⅲ, C Ⅳ in model group increased significantly, serum PCⅢ, HA in Jiajiansanjiasan group and positive control group increased significantly; Compared with model group, serum HA, LN, PC in positive control group and Jiajiansanjiasan group decreased significantly.3. Compared with control group, the IL-1, IL-6, TGF-a in model group increased significantly; the IL-1, IL-6, TGF-α in Jiajiansanjiasan group and positive control group has no significant difference. Compared with model group, the IL-1, IL-6, TGF-a in positive control group and Jiajiansanjiasan group decreased significantly.4. Compared with control group, the TGF-β1 and Smad2 in model group increased significantly. Compared with model group, the TGF- β1 and Smad2 in Jiajiansanjiasan group decreased significantly.Conclusions:1. Jiajiansanjiasan can improve pathological morphology and parts of the serum liver fibrosis markers.2. Jiajiansanjiasan can reduce the IL-1, IL-6, TGF-a, suppression of inflammation, it has function to anti-hepatic fibrosis.3. Jiajiansanjiasan can reduce the expressive level of TGF-β1 and Smad2, the mechanism of its anti-hepatic fibrosis function may be it’s can inhibiting TGF-β1, influnce the TGF-β1/Smad2 signaling pathway.
