Objective :To study the relation of mitochondrial oxidative stress and pμlmonary fibrosis and the productive effects of matrine on the oxidative damage of lung mitochondrial in pμlmonary fibrosis Rats.Methods:Male SD rats randomly divided into five groups, two control groups (one is the normal control group C,another is the bleomycin model group,B) and four test groups intervened respectively with matrine 50mg/kg, matrine 100mg/kg, prednisone,at the same time administered bleomycin.Each group were randomly sacrificed on day 7,14,and 28 respectively.The lung mitochondrial oxidative stress level was determined by measureing the8-hydroxy-2-dexyguanosine of mitochondrial DNAand the activities of fumarase and anconitase.Resμlts:Resμlts with HE staining under light microscope: Capillaries of lung were congested,the breadth of alveolus interval to increase and dropsy,and there are large nunmbers of neutrophil and monocyte were observed,fiber hyperplastic focuses were obseved lying around alveolus interval and bronchiole in B group of 7d. the inflammation of B group of 14d were lighten and showed preliminary finrosis focus.Inflammatory of every group reduced on 28d.Fibrosis degree of B group was more obviously and had a lot consolidation area,this showed the animal model replication was success.Conditions of D group and B group became no more distinctly. Between C group and E group showed significance.Aconitase activities: Compared with B group and C group,the aconitase activities reduced significantly on 14d and 28d.compared with M100 group and B group displayed B group significantly lower aconitase activity than M100 group. Fumarase activities:Mitochondria isolated from the lungs of different groups displayed a similar level of fumarase activity as mitochondria isolated from control mice.Levels of mtDNA 8-hydroxy- 2-deoxyguanosine (8OHdG) in B group mouse lungs were significantly increased compared with control mice on 14d and 28d.Levels of mtDNA 8-hydroxy- 2-deoxyguanosine (8OHdG) in M100 group mouse lungs were significantly lower compared with B group mice.Statistical significance was determined by a Student'st test and isdefined as P < 0.01.Conclusion:1. Bleomycin can induce pμlmonary mitochondria oxidative stress and pμlmonary fibrosis.2. Matrine can obviously reverse the process of pμlmonary fibrosis which may have something to do with the inhibition of the mitochondria oxidative stress.
