Objective: To study the effects of Ningshenling Granule on blood glucose,body weight,behavior and expression of neurotransmitters DA and CaMKII in diabetic hippocampus rats,and to explore the mechanism of Ninglingling Granules in treating diabetes with depression.Methods: A total of 100 SPF male Sprague-Dawley rats were randomly divided into two groups: group I was blank and 10 were given normal feeding,and group II was 90.Group II was fed with high-fat diet and intraperitoneal injection of streptozotocin for replication of the diabetic model,and then replicated in the CUMS model.They were randomly divided into 5 groups(model group,metformin group,metformin + Ningshenling group,metformin + Shugan Jieyu group,metformin + fluoxetine group),and the changes of body weight,blood glucose and behavior were measured.The hippocampus was observed by immunohistochemistry.mRNA expression levels of DA and CaMKII;Real-time PCR method was used to detect protein changes of DA and CaMKII.Result:1.Effects on body weight: After drug intervention,the weight of metformin group was significantly higher than that of model group(P<0.05).Antidepressant group(Metformin + Ningshenling group,metformin + fluoxetine group,metformin + Shugan)There was no significant difference in body weight between the two groups(P>0.05).2.Effects on blood glucose: There was no significant difference between metformin + Ningshenling group,metformin + fluoxetine group,metformin + Shugan Jieyu group and metformin group(P>0.05).3.Behavioral testing: The frequency of activities in metformin + Ningshenling group,metformin + fluoxetine group,metformin + Shugan Jieyu group was higher than that in metformin group(P<0.05).4,D2 mRNA,CaMKII? mRNA expression: Metformin group compared with the model group D2 mRNA,CaMKII? mRNA expression was significantly increased(P <0.05);metformin + Ning Shenling group,metformin + fluoxetine group,metformin + Shugan Jieyu group and There was no significant difference in the metformin group.5,DA,CaMKII expression: metformin + Ningshenling group,metformin + fluoxetine group,metformin + Shugan Jieyu group compared with metformin group DA,CaMKII expression was significantly increased(P <0.05).Conclusion: 1.Ning Shenling granules can increase the number of spontaneous activities and improve the depression behavior of diabetic rats with depression.2.Simple hypoglycemic treatment increased the expression of D2 mRNA and CaMKII? mRNA,but the expression of CaMKII and DA protein did not change.3.Ningshenling Granules can increase the expression of D2 mRNA and CaMKII? mRNA in hippocampus of rats,and increase the expression of CaMKII and DA.
