[Objective]The purpose of this study was to detect APC gene mutations in osteosarcoma tissues and to analyze the correlation between APC gene mutations and clinical pathological characteristics and prognosis of osteosarcoma patients.To evaluate its diagnostic value for osteosarcoma and whether it can be used as a prognostic molecular marker for osteosarcoma,in order to find a gene target that can help to accurately evaluate and improve the prognosis of patients,and to explore a new way for the prognosis evaluation and treatment of osteosarcoma.[Method]APC gene mutations in 30 osteosarcoma tissues and 10 osteochondral tissues were detected by Ion Torrent sequencing technology;Data analysis was carried out by using the tool of Life Technologies' PGM Server Sequencing Analysis Integration;SNV and InDel were processed by the official software AmpliSeq of PGM Server,and VCF results were obtained by SNV and InDel detection,and the results were best after text processing and annotation of annovar software.Final outcome document;Chi-square test(Fisher's exact probability)was used to analyze the experimental results,and to explore the relationship between APC gene mutation and sex,age,location,pathological type,lung metastasis,clinical stage,pathological fracture,alkaline phosphatase level,recurrence and survival status of osteosarcoma patients.Kaplan-Meier and logarithmic rank test were used to analyze the relationship between the mutation of the APC gene and the 5-year overall survival rate.The survival function curve was drawn by GraphPad prism6.0 software.[Result]1.In 30 osteosarcoma tissues and 10 osteochondral tissues,one mutation site of APC gene was RS5 112 175769,which was synonymous with exon mutation.2.APC gene mutation was detected in 20 samples of 30 osteosarcoma tissues,and the total mutation rate of APC gene was 66.67%.APC gene mutation was detected in 2 samples of 10 osteosarcoma tissues,and the total mutation rate of APC gene was 20%.The mutation rate of experimental group was significantly higher than that of control group(P < 0.05).3.There was significant difference between APC gene mutation and pulmonary metastasis in osteosarcoma patients(P < 0.05).There was no significant difference in mutation rate of Rs5 112175769 locus in sex,age,tumor location,pathological type,clinical stage,pathological fracture,alkaline phosphatase level and recurrence of osteosarcoma patients(P > 0.05).4.TThe mutation rate of APC gene was 59.1% in survival group and 87.5% in death group,with statistical significance(P < 0.05).5.The 5-year survival time of APC gene mutation patients was significantly different from that of non-mutation patients(P < 0.05).[Conclusion]The mutation rate of APC gene in osteosarcoma patients was significantly higher than that in osteochondroma control group,which may be one of the causes of osteosarcoma.The mutation of APC gene Rs5 112175769 was not related to clinical and pathological factors,but only related to lung metastasis.APC gene mutation is unfavorable to the survival and prognosis of osteosarcoma patients,and may be a marker for evaluating the adverse prognosis of osteosarcoma patients.
