?Objective? Many studies have confirmed that the occurrence and development of tumors are often associated with mutations in certain related genes.In recent years,gene detection techniques have been gradually applied to the treatment of cancer patients.In this study,22 cases of osteosarcoma and 10 cases of osteochondroma tissue samples were used to test about the mutation of ERBB4 gene that based on the Ion Ampli Seq ? Cancer Hotspot Panel V2.And the clinical and pathologic factors related to osteosarcoma patients were analyzed between different groups,with a view to the prognosis of patients with osteosarcoma and its treatment to explore new ways.?Methods? 1.Collect samples.Extract DNA from 22 cases of osteosarcoma tissue and 10 cases of osteochondroma tissue.2.Enriching target DNA fragments follows the protocol of Life Technology Cancer Panel,and sequencing after purification and library.3.Sequencing data analyse and get the variations.Based on the clinical data collected,to analyze the relationship between ERBB4 mutation rate and the age,sex,tumor location,clinical Enneking stage,recurrence and pulmonary metastasis,pathological type,alkaline phosphatase level by statistical method.And to analyze the relationship between ERBB4 gene mutation and 5-year survival rate of osteosarcoma patients?Results? The results of Ion Torrent sequencing showed that ERBB4 gene mutations occurred in 14 cases of experimental group(osteosarcoma group).The mutation rate was 63.6%.And there was no mutation of ERBB4 gene detected in control group(osteochondroma group).The difference was statistically significant(P < 0.05).Of the 10 patients with lung metastases,the ERBB4 gene mutation was detected in 9 patients with a mutation rate of 90%,which was significantly higher than 41.7%(P < 0.05)in patients without lung metastases.There was no significant difference(p > 0.05)about ERBB4 gene mutation rate in other clinical and pathological factors subgroups such as the age,sex,tumor location,Enneking surgical staging,pathological fracture,alkaline phosphatase elevation,recurrence and pathological type subgroup.Survival function curves show that ERBB4 mutations are associated with poor overall survival rates(p <0.05).?Conclusion? The rate of ERBB4 gene mutation in osteosarcoma tissues is high.ERBB4 gene may play a role in the occurrence and development of osteosarcoma.ERBB4 gene mutation may be a poor prognosis of osteosarcoma patients.And ERBB4 may become a potential target of gene therapy and targeted therapy of osteosarcoma patient.
