| Objective In this study,the mutation of LKB1 gene in osteosarcoma cells was detected,combined with the detection results to analyze the correlation between clinical and pathological factors related to osteosarcoma patients,and provide a prognostic and gene-targeted therapy for osteosarcoma patients.Theoretical feasibility.Methods 1.Select osteosarcoma tissue samples(classical osteosarcoma)from 28 patients with primary osteosarcoma undergoing standardized surgery between January 2010 and May 2017 at the Second People’s Hospital of Shenzhen City,Department of Bone Tumors,and from the organization DNA was extracted from the specimen.2.According to the Life Technology Cancer Panel standard technical procedure,multiplex PCR enrichment was performed on the extracted DNA samples,and the resulting DNA fragments were purified and constructed and then sequenced on an Ion Torrent sequencer.3.Combined with the clinical data of the patients collected,the use of SPSS 19.0 software and Fisher’s exact probability method to investigate the mutation of LKB1 gene and the patient’s age,gender,tumor location,clinical Enneking stage,recurrence and lung metastasis,pathological type and alkaline Correlativity between phosphatase levels,Kplan-Meier and log-rank tests were used to further analyze the relationship between LKB1 gene mutations and 5-year survival in patients with osteosarcoma,and GraphPad Prism 6.0 software was used as a survival function curve.Results 1.A total of 17 cases of LKB1 gene mutations were detected in 28 specimens of osteosarcoma,and the mutation rate was 60.71%.There are two common mutation sites in all specimens,that is,mutations at positions 122031 and 1223125 of chromosome 9 occur,and mutation at position 122031 changes from base T to base C.The mutation site is located in the intron region,and the mutation at position 1223125 is a mutation from base C to base G.This mutation is located in the exon region.The reliability of the above two mutation sites under the GATK(second generation sequencing analysis software)statistical analysis of their mutations were: 122031 site mutation false positive is low,the mutation is reliable;and 1223125 site mutation false positive Higher,the mutation is not reliable,be removed.2.Among the 18 patients with pulmonary metastases,15 cases of LKB1 gene mutation occurred,and the mutation rate was as high as 83.3%.However,4 out of 10 patients without pulmonary metastases had mutations.The mutation rate was 40.0%.3.The mutation rate of LKB1 gene in patients with gender,age,tumor location,pathological fractures,elevated alkaline phosphatase,Enneking surgical stage,recurrence and pathological subtypes of the subgroup between the mutation rate was no significant difference(p> 0.05),survival The function curve shows that the LKB1 mutation is associated with a five-year survival rate in osteosarcoma patients(p<0.05).Conclusion LKB1 mutation may be related to metastasis in lungs in patients with osteosarcoma;LKB1 gene mutation may be a factor for poor prognosis in patients with osteosarcoma;LKB1 gene may become a potential target for gene therapy and targeted therapy of osteosarcoma. |
