Gastric cancer is one of the most common malignant tumors of digestive tract.Its incidence ranks fourth and its mortality ranks third among all malignant tumors in the world.Although the global incidence of gastric cancer has declined in recent years,its threat to human health should not be underestimated.Compared with the traditional treatment methods,targeted therapy has specificity and selectivity,which greatly reduces the toxicity and side effects on normal cells and tissues.Therefore,it is particularly important to find new targets for early diagnosis and treatment of gastric cancer.Fibronectin Type ? Domain Containing 1(FNDC1)contains Type ? fibronectin domain(FN-?),which constitutes one of the FN1 domains,and FN1 can regulate cell migration,proliferation and apoptosis.At the same time,studies have shown that FNDC1 can promote vascular endothelial growth factor(VEGF)to induce angiogenesis.The expression level of FNDC1 in prostate cancer tissue is significantly higher than that in normal tissue.However,the expression of FNDC1 in gastric cancer or other tumors is still not clear,we put forward a hypothesis that whether FNDC1 plays an important role in the occurrence and development of gastric cancer.Therefore,this research intends to study the expression of FNDC1 in gastric cancer tissues and adjacent normal tissues,and to analyze the relationship between FNDC1 expression and clinicopathological characteristics and prognosis of gastric cancer patients,thus providing reference for the diagnosis and treatment of gastric cancer.Purpose: This study was aimed to measure the expression level of fibronectin Type ? domain containing 1(FNDC1)in patients with gastric adenocarcinoma(GAC)and to evaluate its prognostic value.Methods: We used bioinformatics prediction and immunohistochemical validation to examine the expression level of FNDC1 in GAC.Ninety cases of tumor tissue samples and paired adjacent normal tissues were retrospectively collected to detect the expression of FNDC1 by immunohistochemical staining.Besides,chi-square test,Kaplan-Meier method,log-rank test and multivariate Cox regression survival analysis were performed to identify the correlation between the FNDC1 expression and pathological parameters of GAC patients and its prognostic significance.Results: The data from Oncomine and TCGA predicted that the expression level of FNDC1 was significantly higher in GAC tissue than that in normal tissues(all P<0.05).Further survival analyses indicated that the overall survival(OS)and disease-free survival(DFS)of patients with high expression of FNDC1 was shorter than that of patients with low expression of FNDC1(all P<0.05).Immunohistochemical results showed that the expression level of FNDC1 protein in GAC tissues was higher than that in normal tissues(P=0.011).And the overall survival(OS)of patients with high FNDC1 expression level was shorter than that of patients with low FNDC1 expression level(P=0.002).Meanwhile,Kaplan-Meier univariate survival analysis and cox multivariate survival analysis revealed that high FNDC1 expression level was independent risk factor for GAC patients(P=0.002 and P=0.048).Conclusion: The study confirmed that FNDC1 was highly expressed in GAC patients,and the overexpression of FNDC1 was closely related to the poor prognosis of GAC.
