Background and ObjectiveThe protein encoded by PIK3C2 A gene belongs to the phosphoinositide3-kinase(PI3K)family.PIK3C2 A plays an essential role in angiogenesis,abnormal expression of PIK3C2 A has been reported in several cancers such as breast cancer,cervical cancer,lung cancer,stomach cancer,colon cancer,liver cancer and so on.PI3-kinases is a foundational function in many important signaling pathways,such as cell proliferation,cell migration,intracellular protein trafficking,cell survival,and oncogenic transformation.Our previous pilot study on profiling gene expression in peripheral blood of AMI patients indicated that AMI patients had a low expression of PIK3C2 A.This study aims to make an assessment on whether expression of PIK3C2 A gene can be used as a biomarker for predicting the risk of acute myocardial infarction.By means of experiments and systematic analysis of the data,the further research on the role of PIK3C2 A gene in acute myocardial infarction.MethodsWe collected peripheral blood from 84 subjects with non-coronary heart disease(non-CHD)and 70 patients with AMI.We analyzed and compared the clinical data of non-CHD patients and AMI patients.All the patients were given a CAD severity score by Gensini scoring system.The real-time quantitative PCR test was applied to measure levels of PIK3C2 A gene expression at m RNA level in peripheral blood.ResultsThere was not significant difference between the AMI group and thenon-CHD group in terms of Serum triglyceride level(TG),serum total cholesterol level(TC),high-density lipoprotein cholesterol(hdl-c)and low-density lipoprotein cholesterol(ldl-c),BMI,gender,smoking history,history of hypertension,history of diabetes,family history of coronary heart disease,systolic blood pressure,diastolic blood pressure.The age of patients in the acute myocardial infarction group was significantly higher than that in the non-coronary heart disease group,and the difference was statistically significant(t=-3.133,P=0.002).The fasting blood-glucose of patients in the acute myocardial infarction group was significantly higher than that in the non-coronary heart disease group,and the difference was statistically significant(z=-2.899,P=0.004).There was no statistically significant difference between the patients in the acute myocardial infarction group and the non-coronary heart disease group in the administration of aspirin,ticagrelor and hydroclopidogrel after admission,while the rate of using anticoagulant administration in the acute myocardial infarction group was significantly higher than that in the non-coronary heart disease group,with a statistically significant difference(x~2=72.894,P<0.001).In peripheral blood of the acute myocardial infarction Group,the level of PIK3C2 A m RNA was significantly lower than that of the non-coronary heart disease group(z=-2.606,P=0.012).The relative expression quantity of PIK3C2 A gene in the acute myocardial infarction Group Group was 0.85 times of that in the non-coronary heart disease Group.The relationship between the clinical data and the expression level of PIK3C2 A gene was further analyzed.Results: the relative expression level of PIK3C2 A gene m RNA was different between the older group and the younger group(z=-2.648,P=0.008).The relative expression of PIK3C2 A m RNA level was not related to fasting blood glucose level(z=-1.621,P=0.105).The relative expression level of PIK3C2 A gene m RNA was notrelated to the administration of anticoagulant drugs(z=-1.449,P=0.147).Binary Logistic regression analysis indicated that low expression of PIK3C2 A was closely related with AMI and the OR value was 2.231.Fasting blood-glucose also was closely related with AMI and the OR value was 3.674.Pearson correlation analysis between relative expression quantity of PIK3C2 A gene and severity of coronary artery lesion,cardiac troponin I showed that the expression quantity of PIK3C2 A at the m RNA level was not correlated to Gensini score(r_s=0.017,P=0.892)and serum Tn I concentration(r_s=0.103,P=0.429).ConclusionThe level of PIK3C2 A gene expression in patients with AMI is significantly lower than that of non-CHD people.Low expression of PIK3C2 A gene is an independent risk factor of AMI.Low expression of PIK3C2 A could serves as a potential biomarker to predict risk of AMI.
