Study On The Effect Of Hongsiwu Decoction Combined With Effective Ingredients On Early Callus Healing And Its Mechanism In Rats

Objective: Through imaging(Micro-CT),histopathology(HE staining),vascular endothelial growth factor(VEGF)concentration determination,joint swelling,joint analysis to measure peach siwu decoction prescription drugs main effective monomer components compatibility to the influence of early fracture healing in rats and the possible mechanism,for the rich peach siwu decoction prescription drugs main effective monomer components compatibility in the clinical application provides the basis.Methods: Sixty normal SD rats were selected,male,with a body weight of(200 20g).After 1 week of adaptive feeding,the animals were randomly divided into normal group,sham operation group,model control group,original prescription water extract group and monomer component prescription group,with 12 in each group.The left open femoral fracture model of rats was established in a sterile environment,and it was required to be constructed(model control group,original formula water extraction group,monomer component formula group).The losers in the operation were removed and added to the number required in this experiment.After the operation,the original prescription water extract group was given peach siwu decoction,the monomer component group was given peach siwu decoction,the main monomer component group was given peach siwu decoction,and the other three groups were given the same amount of normal saline.Mold was made in fracture treatment respectively the fourth day,7 days,10 days after three phase points record changes of body weight of rats and joint swelling degrees,and by observing the rat serum vascular endothelial growth factor(VEGF)concentration,Micro-CT,histopathology and rats joint swelling degree comprehensive explore peach siwu soup main effective monomer composition composition influence on the early fracturehealing in rats,to study the possible mechanism.Result: 1.The degree of limb swelling can be detected in rats,and the degree of swelling in the water extract group and monomer component group decreased significantly faster than that in the model group at 4,7 and 10 days after surgery(P <0.05).There was no significant difference in swelling degree between the monomer component group and the original formula group(P > 0.05).2.Serum VEGF concentration was significantly higher in the water extract group and monomer component group than in the model group at 4,7 and 10 days after surgery(P < 0.05).There was no significant difference in VEGF concentration between the monomer component group and the water extract group(P > 0.05).3.Callus bone mass 4,7 and 10 days after surgery,callus bone mass in the original water extract group and monomer component group was significantly higher than that in the model group(P < 0.05).There was no significant difference in callus bone mass between the monomer component group and the original water extract group(P >0.05).4.Bone callus volume ratio on the 4th,7th and 10 th days after surgery,the volume ratio of bone callus between the original water extract group and the monomer component group was significantly higher than that of the model group(P < 0.05).There was no significant difference in callus volume ratio between the monomer component group and the original water extract group(P > 0.05).5.The postoperative activity of the original recipe with water extract and monomer component was faster than that of the model group.Conclusion: 1.The compatibility of the main effective monomer components in the prescription of taohong siwu decoction can promote the open fracture healing of rats and shorten the healing time of fractures.2.There is no significant difference between the compatibility of main effective monomer components in the prescription of taohong siwu decoction and the original decoction in the treatment of open fractures.
