Objective:Through the PI3K/Akt signaling pathway of network pharmacology,to explore the possible mechanism of action of Taohong Siwu Decoction,a representative prescription for promoting blood circulation and removing blood stasis,in intervening in the early stage of rat fracture model.Methods:The first part is to find the potential material basis and related signal pathways of Taohong Siwu Decoction intervention in the early stage of femoral fracture through network pharmacology.The second part of animal experiments continued to verify.50 healthy SD male rats weighing(200 ± 20g)were randomly divided into normal group,sham operation group,model group,medicinal soup group and control group,with 10 rats in each group.Establish rat femoral fracture models for the model group,drug group and control group.After the operation,the drug group was given Taohong Siwu Decoction by gavage,the control group was given Jiegu Qili Capsules with distilled water by gavage,and the other three groups were gavage with the same amount of normal saline twice a day for a total of 10 days.By observing the general conditions of the rats,histopathology(HE staining),immunohistochemistry(ELISA)and other methods,explore the possible mechanism of Taohong Siwu Decoction intervention in the early stage of the rat fracture model.Results:1.Through the method of network pharmacology,find the PI3K/Akt signal pathway which is closely related to the early stage of femoral fracture.2.Taohong Siwu Decoction significantly improved the general activity state of the rat fracture model in the early stage,and improved the clinical symptoms of the rat to a certain extent,and no adverse reactions were found.It is safe to take it under the conventional dosage.3.The HE staining results showed that a small amount of chondrocytes and granulation tissue were seen in the model group,but no bone trabecula formation;a large number of chondrocytes and granulation tissues were seen in the Taohong Siwu Decoction group and the Jiegu Qili group,and a small amount of bone Beam formed.4.Serum phosphoinositide 3-kinase(PI3K)concentration and phosphorylated protein kinase B(p-Akt)concentration,Taohong Siwu Decoction group and Jiegu Qili group are higher than the model group(P<0.05),Taohong Siwu Decoction There was no significant difference between the Jieguqili group and the Jieguqili group(P>0.05).5.Serum VEGF concentration,the VEGF concentration of Taohong Siwu Decoction group and Jieguqili group were higher than the model group,there was no significant difference between Taohong Siwu Decoction group and Jieguqili group(P>0.05).Conclusion:1.The early intervention mechanism of Taohong Siwu Decoction on fracture may be through regulation of PI3K/Akt signaling pathway.2.Taohong Siwu Decoction effectively promotes the early healing of rat fractures and improves the efficiency of fracture healing.
