Objective:To establish the relationship between fingerprints and antitussive effect and to Clarify the material basis of its cough-relieving effect by analyzing the relationship and serum pharmacochemistry of total alkaloids from Stemona tuberosa Lour.from 29 producing areas in China.Metheods:1.It is analyzed that HPLC chromatogram of total alkaloids from Stemona tuberosa Lour.from 29 producing areas in China.2.Pharmacodynamic experiments on antitussive effects of total alkaloids from Stemona tuberosa Lour.were conducted with cough times,cough suppression rate and cough incubation period of mice within 2 min as efficacy evaluation indexes.3.Using grey correlation analysis method,correlation analysis was carried out on the 8 common peak areas of HPLC chromatogram of total alkaloids from Stemona from 29 producing areas in China and 2 evaluation indexes?cough times within 2 min,cough suppression rate?for cough relief,and the spectral-effect relationship of antitussive effect of total alkaloids from Stemona tuberosa Lour was studied.4.Using partial least squares regression analysis method,the correlation analysis was carried out between the 8common peak areas of HPLC chromatogram of total alkaloids from Stemona tuberosa Lour.from 29 producing areas in China and 2 evaluation indexes?cough times within 2 min,cough suppression rate?for cough relief,and the spectrum-effect relationship of antitussive effect of total alkaloids from Stemona tuberosa Lour.samples collected in 29 producing areas in China was studied.5.UPLC-Q-TOF-MS and Metabolynx analysis methods were used to study the serum pharmacochemistry of total alkaloids from Stemona tuberosa Lour.Results:1.There are 8 common peaks in HPLC chromatograms of total alkaloids from Stemona tuberosa Lour.from 29 producing areas in China.2.The total alkaloids of Stemona tuberosa Lour.significantly inhibit the ammonia induced cough in mice,showing the consistency of antitussive action of Radix Stemonae.3.The grey relational analysis shows that the order of correlation degree x0 between common peaks of total alkaloids in Stemona tuberosa Lour.and cough times is Oxytuberstemonine>Isotuberostemonine>Croomine>No.7>Tuberostemonine>No.8>Tuberostemonine H>Neotuberostemonine,and the order of correlation degree x0 between common peaks and cough suppression rate is No.4>Croomine>Tuberostemonine H>Neotuberostemonine>Isotuberostemonine>Oxytuberstemonine>No.7>No.8;4.The partial least square regression equation between the cough times of the mice within 2 minutes and HPLC chromatogram of total alkaloids of Stemona tuberosa Lour.is YA=-0.07X1-X2+0.75X3-0.12X4-0.11X5+0.38X6-0.31X7-0.07X8?YA is the coughing times of mice within 2min after gavage administration of total alkaloids of Stemona tuberosa Lour.,Xn is the contribution value to the common peaks of total alkaloids of Stemona tuberosa Lour.?;The partial least square regression equation between cough suppression rate and HPLC chromatogram of total alkaloids from Stemona tuberosa Lour is YB=0.01X1+0.05X2-0.77X3-0.09X4+0.09X5-0.28X6+0.25X7+0.03X8(YB is coughing suppression rate after gavage administration of total alkaloids from Stemona tuberosa Lour.to the mice,Xn is contribution value to each common peak of total alkaloids from Stemona tuberosa Lour.4.Fifteen blood-entry components were found in the medicated serum of total alkaloids from Stemona tuberosa Lour.,which were presumed to be the new ring-opening derivatives of Neotuberostemonine and metabolites of ineotuberostemonol after analysis.Conclusion:1.The HPLC fingerprints is related to cough antitussive effect of total alkaloids from Stemona tuberosa Lour.samples collected in 29 areas in China.The relationship between fingerprints and antitussive effect of total alkaloids from Stemona tuberosa Lour.shows that Croomine and No.7unknown alkaloid may be the material base for relieving coughing.2.The new ring-opening derivatives of Neotuberostemonine and metabolites of ineotuberostemonol they may be the material basis of antitussive effect of total alkaloids from Stemona tuberosa Lour.
