Objective:1.By simulating the reality of military transport operations time,we establish an acute rats liver injury model in a-10℃freezer environment.2.To study the mechanism of acute liver injury induced by cold stimulation.3.To work out the best medication plan by comparing the efficacy of Chaihu Guizhi Gan Jiang Decoction high-dose group,low-dose group and conventional dose Silibin Meglumine Tablets in the study.Methods:1.Establishing an acute rats liver injury model in a-10℃freezer environment to observe the rat liver function(ALT,AST)and pathological conditions exposed to low temperature of 2h,4h,6h,and the success time of modeling was determined.2.Comparing the efficacy of Chaihu Guizhi Gan Jiang Decoction high-dose group,low-dose group and conventional dose Silibin Meglumine Tablets,and to study the mechanism of acute rats liver injury induced by cold stimulation by detecting the serum levels of ALT,AST,H2O2,GPx and TNF-α,IL-10,Bcl-2,Bax,and to determine the optimal dose that can effectively prevent and treat acute liver injury induced by cold stimulation.Results:1.After low temperature exposure for 2 hours,4 hours,and 6 hours,the levels of ALT and AST increased significantly in the model group compared with the normal group at 4 hours(P<0.01).The experimental datas showed that the model was successful after 4 hours cold exposure in rats.After 6 hours cold exposure,the rat liver function still maintained a degree of liver injury.2.After 7 days of preventive treatment,(1)In improving liver function,the levels of ALT and AST in the 4 hours group were lower than those in the model group,but significantly lower in the high dose group and silymarin group(P<0.05).The levels of ALT and AST in the 6h group were lower than those in the model group,and it was significantly lower in the high-dose group(P<0.05);(2)In terms of anti-oxidant,Compared with the treatment group,the level of H2O2 in the 4 hours group was significantly lower in the three groups,the significantly lower in the high-dose group and silymarin group(P<0.01).The level of GPx in the 4 hours treatment group was significantly higher than that in the model group(P<0.01).The level of H2O2 in the 6 hours group was significantly lower in the three treatment groups,and it was significantly lower in silybin group(P<0.05),and the level of GPx in the 6 hours group was increased to some extent in the treatment group,and it was significantly increased in silymarin group(P<0.01).(3)In terms of anti-inflammatory factors,the level of TNF-αin the 4 hours group was decreased in the treatment group,and significantly lower in the high-dose group and silymarin group(P<0.05).In the IL-10 level of the rats,IL-10 levels increased in the treatment group,and the high-dose group increased significantly(P<0.01),silybin group(P<0.05);The TNF-α,IL-10 levels of 6 hours group,was no significant difference among the treatment groups(P>0.05).(4)Anti-cell injury and apoptosis,the Bax level in the 4 hours group was decreased in all treatment groups compared to the control group,and the low dose group and high dose group had significant statistical significance(P<0.01),but silybin group(P<0.05);Bcl-2 levels in 4 hours group was increased in each group compared to the model group,and increased significantly in the high-dose group(P<0.05);Bax levels in the 6 hours group were decreased in all groups,with statistical significance in the low-dose group and high-dose group(P<0.05);Bcl-2 in the 6 hours group was increased in each group,but no significant statistical significance(P>0.05).Conclusion:1.The experiment is easy to operate,and the rats are placed in the low-temperature freezer to build models with controllability and cost savings;2.Chaihu Guizhi Gan Jiang Decoction as the empirical formula of"Treatise on Febrile Diseases",is effective in preventing acute liver injury in rats induced by cold stimulus.And it can improve liver function,anti-oxidation,reduce the production of inflammatory factors,and can reduce the production of rat liver apoptotic proteins.The high-dose of Chaihu Guizhi Gan Jiang Decoction has advantages in preventing and treating liver damage,and its gas between Silibinin groups is not obvious.
