Objective:(1)To establish an model of acute kidney injury induced by underwater explosion in rabbits and to provide a basis for further study.(2)According to this model,the concentrations of serum NGAL,KIM-1 and MMP-9 were tested by ELISA and the expression of mmp-9 in kidney was also analyzed by Western blot.The purpose of this article is to study the significance of these factors in rabbits with acute kidney injury caused by underwater explosion.Methods:(1)40 New Zealand rabbits were randomly divided into 4 groups: the normal group(N1 group),the 1.8m group,the 1.0m group and the 0.5m group,10 cases in each group.After the rabbits were anesthetized,the explosion was carried out in three experimental groups by using 1.0g taian as explosive source.After explosion,the survival rate,physiological parameters and the pathological changes were recorded,the dynamic variations of serum NGAL in different periods were tested through ELISA.(2)Useing this model,20 rabbits were divided into normal group(N2 group)and injury group(I group)randomly,10 cases in every group.I group was developed by using 1.0 g taian as an underwater explosive source,after the explosion,simple treatment was carried out.The dynamic variations of NGAL?KIM-1 and MMP-9 were tested through ELISA,the MMP-9 in the kidney tissue was evaluated by Western blot.Results:(1)In general,the N1 group survived after 24 h,the pathological result of glomerulus,renal tubule and renal interstitial structure was clear.In the group of 1.8m and 1.0m,the death rate was 10%,pathological analysis showed focal hemorrhage in 1.8m while more hemorrhage in the renal interstitial structure and the infiltration of inflammatory cells in 1.0m.The group of 0.5m,the early death rate was 20%,while it increases to 50% after 13 h.Compared with the normal group,the serum NGAL concentration started to rise(p < 0.05)after 4h,and the 24 h was still statistically significant.(2)After AKI caused by the underwater explosion,the concentrations of NGAL and MMP-9 in I group were significantly increased as compared with the N2 group(P<0.01),however,the KIM-1 was still no statistically significant difference(P>0.05),Western blot showed that the MMP-9 was hardly expressed in N2 group,while the expression of MMP-9 was significantly increased in I group.Conclusion:(1)The group of 1.0m can simulate the effect and provides an underwater blast injury model of AKI in rabbits.(2)We conclude that the serum NGAL or MMP-9 could be early biomarkers of AKI caused by underwater explosion.
