Objective To explore early changes of neutrophil gelatinase associated lipocalin(NGAL) and kideny injury molecule-1 (KIM-1) in lipopolysaccharide(LPS)-induced acute kidney injury (AKI) in rats.Methods Thirty-two male Wistar rats were randomized to receive caudal injection of 1 ml normal saline (control group), or 5 mg/kg LPS (LPS2h,4h,6h group). After the prescribed time, the blood and urine levels of NGAL and the level of urine and nephridial tissue of KIM-1 were determined with enzyme-linked immunosorbent assay (ELISA), serum creatinine(Scr) and urine creatinine (Ccr) with Jaffe's reaction and the morphology of kidneys observed under the optical microscope.Results Compared with normal control group, the level of Scr was elevated slightly in the LPS2h group (P>0.05), significantly in the LPS4h,6h group (P<0.01); the level of Ucr was decreased slightly in the LPS2h group (P>0.05), significantly in the LPS4h,6h group (P<0.01); the levels of the serum and urine of NGAL were elevated significantly in the LPS2h,4h,6h group (P<0.01); the levels of urine and nephridial tissue of KIM-1 were elevated significantly in the LPS2h,4h,6h group (P<0.01). Compared with LPS2h group, the level of Scr was elevated significantly in the LPS4h,6h group (P<0.01); the level of Ucr was decreased significantly in the LPS4h,6h group(P<0.01); the levels of the serum and urine of NGAL were increased slightly in LPS4h group (P>0.05), significantly in the LPS6h group (P<0.01); the levels of urine and nephridial tissue of KIM-1 were elevated significantly in the LPS4h,6h group (P<0.05, P<0.01); Compared with LPS4h group, the level of Scr was elevated significantly in the LPS6h group (P<0.01); the level of Ucr was decreased slightly in the LPS6h group (P>0.05); the levels of the serum and urine of NGAL were increased significantly in LPS6h group (P<0.01); the levels of urine and nephridial tissue of KIM-1 were elevated significantly in the LPS6h group (P<0.01).Conclusion NGAK and KIM-1 were increased in the early period of AKI and may be used as a marker for early diagnosis; the KIM-1 had time dependence and may be used as a monitor marker of AKI.
