Objective: 1.The expression of Kiss-1 and Notch-1 gene in breast cancer tissues,adjacent normal tissues and benign tumor tissues of breast cancer was studied.2.Comparing the expression of Kiss-1 and Notch-1 with patients with different clinical conditions,such as age,tumor size,TNM stage and lymph node metastasis.3.To investigate whether the expression of Kiss-1 and Notch-1 in cancer tissues is interrelated.Methods: 1.The selection of our college in May 2016 to February 2016 surgical oncology ward 2,30 cases of clinically diagnosed breast cancer patients take the cancer tissue and normal tissue adjacent to carcinoma(cancer selected are from normal tissue adjacent to carcinoma tissue and more than 5 cm is not in the same quadrant,by pathological diagnosis cut edge no cancer cells),and then take 20 cases of breast benign tumor,benign tumor of breast fibroadenoma)organization.2.Real-time reverse transcription fluorescence quantitative PCR(qrt-pcr)was used to determine the expression of the Kiss-1 gene and the Notch-1 gene in cancer tissues,adjacent normal tissues and benign tumor tissues at the mRNA level.3.Immunohistochemical(SP)technology was used to verify the gene expression of cancer tissues,adjacent normal tissues and benign tumor tissues from the protein level again.4.Collected and sorted out the clinical pathological data of patients,and analyzed the relationship between the expression of the above genes and the development of breast cancer by statistical methods.Results: 1.In qrt-pcr,the expression rate of Kiss-1 mRNA in cancer tissues was 26.7%,and the expression rate of normal breast tissue was 73.3%,and the expression rate of benign tumor was 70.0%.The expression of Kiss-1mRNA in cancer tissues was significantly lower than that of benign lesion tissue(P < 0.05)and normal breast tissue(P < 0.05).Stage for ? and ? period in tumor tissue specimens,the positive rate was significantly higher in installment for ? stage tumor specimens(P < 0.05),no lymph node metastasis of carcinoma tissue cases the positive expression rate is significantly higher than cases with lymph node metastasis(P < 0.05).The results of positive expression were not related to other factors such as tumor size(P BBB 0.05).2.The expression rate of Notch 1 mRNA in cancer tissues was 63.3%,and the expression rate of normal breast tissue was 36.7%,and the expression rate of benign tumor was 20.0%.The expression of Notch-1mRNA in cancer tissues was significantly higher than that of benign lesion of mammary gland(P < 0.05)and normal breast tissue adjacent to cancer(P < 0.05).The clinical stage for ? and ? stage breast cancer,cancerous tissue in the positive rate of Notch-1 significantly below ? stage cases(P < 0.05);In the lymph node metastasis,the positive expression rate of Notch-1 in the metastatic cases was significantly higher than that in the non-metastatic cases(P < 0.05).The results of positive expression were not related to other factors such as tumor size(P BBB 0.05).3.In immunohistochemistry,the results of the two methods were basically consistent.The positive rate of Kiss-1 and Notch-1 in cancer tissues was slightly increased,while the positive expression rate of Kiss-1 was 30%,and the positive expression rate of Notch-1 was 70%.4.The expression of Kiss-1 and Notch-1 in cancer tissues was statistically significantly negative.Conclusion: 1.Kiss-1 gene expression in the cancer tissue was lower than that in normal tissue adjacent to carcinoma and benign tumor tissue,the later and with lymph node metastasis and clinical stage of the cases,the expression rate reduce,further prompt the gene transfer is closely related with breast cancer,may inhibit breast cancer metastasis.2.The Notch-1 gene expression was significantly higher in the cancer tissue and normal tissue adjacent to carcinoma benign tumor,and along with the progress of clinical stages and lymph node metastasis of breast cancer,its expression rate is increased further,prompt the gene transfer also is closely related with breast cancer,may promote breast cancer metastasis.3.The gene that is related to the metastasis of breast cancer by Kiss-1 and Notch-1 has a significant negative correlation with the expression of cancer tissue,suggesting that there may be some kind of association between the two,which may play a synergistic role.
