Objective:The high fat diet and streptozotocin(STZ)induced obese type 2 diabetic rat model,to evaluate the effects of liraglutide on the thickness of epicardial adipose tissue and carotid artery intima-media by using color doppler ultrasound,and to study the effects on Ghrelin expresion.Method:50 male SD rats were selected and divided into one group of 10 rats(namely A group)and one group of 40 rats(namely B group);A group received general diet of rats as control group,without intervention;B group rats were fed with high fat diet 3 weeks,then given streptozotocin 45mg/Kg dose abdominal subcutaneous injection 1 times,after 72 hours,B group of tail vein blood were measured fasting blood glucose,if in the fasting blood glucose level is higher than the standard,indicates that the rat model was established successfully.Then the B group was randomly divided into 3 groups,,with the principle of grouping,the groups were given different doses of liraglutide on rats and intervention,according to the given different dose,the groups werenamed low dose group C(n=13,liraglutide,50ug/kg)and high dose group D(n=13,D,liraglutide 200ug/kg),and the high fat control group E group.The A Group continued to give before,the other groups were fed with high fat diet;all rats were given subcutaneous injection of vitamin D3 needs(4 × 105U/Kg,give drug 1 times every week,3 weeks of continuous injection).Liraglutide,7:30am injection 1 times per day.A group and E group were treated with physiological saline as blank control.Continuous feeding and drug intervention 12 weeks,then fasting for 1 nights,weight and fasting blood glucose tseted the second day.Then rats anesthetized with 10%chloral hydrate intraperitoneal,deteced the thickness of ratepicardial adipose tissue and carotid intima-media by ultrasound,applied automatic biochemical to analyzed fasting blood glucose,blood lipid indexes after(mainly the concentrations of triglyceride and total cholesterol,LDL),and elisa used to inspected blood,fasting insulin concentration,serum ghrelin level using the same method.The expression of ghrelin in islet of rats in different groups was detected by Western blot.Result:1.Compared with than A group,the fasting insulin,blood glucose and blood fat index of E group were significantly higher;compared with group E,the index of C group and D group also had obvious differences,;C group than in D group decreased significantly.Specific values can be shown in table 1.2.The changes thickness of epicardial adipose tissue and carotid intima-media and weight,compared with A group,the datas of E group increased,;compared with E group,C group and D group were decreased;C group than D group of epicardial adipose tissue and carotid intima-media thickness decreased significantly.3.Compared with group A of serum ghrelin level,the E group decreased,and the group C and D higher than E group;C group significantly higher than that in D group.4.The expression of ghrelin in islet:Ghrelin detection results in the E group,as compared with A group,E group was significantly decreased;compared with E group,group C and D of ghrelin increased;Ghrelin of group C increased more than D group.Conclusion:1.The liraglutide peptides not only can reduce blood sugar,blood fat,improve insulin resistance,can also reduce thickness of the epicardial adipose tissue and the carotid artery intima,and which dependent on the dose.2.After the obse type 2 diabetes mellitus be treated with liraglutide,the ghrelin express in thd plasma and the pancreas higher than the control group.
