Objective:Aim of this study is to explore the expression of AFP in placenta and fetal liver of pregnant women with maternal serum alpha-fetoprotein(AFP)during pregnancy and to analyze their correlation.Methods:Subjects were singled out according to the results of the inclusion and exclusion criteria and placental histopathological examination.The subjects were divided into three groups:thecontrol group-unplanned pregnancy,the experimental group 1-serum level of AFP?2.5MoM and Adverse pregnancy outcome group and the experimental 2-normal serum of AFP(0.5?2.499 MoM)but Adverse pregnancy outcome.The samples were taken from Reproduction of department of gynaecology first people's hospital in yunnan province,and all samples of the sick data and prenatal screening were recorded using Excel spreadsheets.Experiment was performed by q-PCR,immunohistochemistry and HE staining,Statistical analysis were using SPSS,GraphPad Prism5.0 and image-pro software.Continuous variables comparison were compared by One way-ANOVA test.A P value of<0.01 was considered to indicate statistical outstanding significant.The AFP mRNA and AFP protein levels of experimental group 1,the experimental group 2 and the control group were compared.Results:1.The comparision of placental histopathological analysis and placental AFP(alpha-fetoprotein)and ?-HCG(?-human chorionic gonadotropin)mRNA between different way of labor induction shows that low-dose rivanol and mifepristone have minimal effects on the maternal surface of placenta(P>0.05);2.The results of healthy placental q-PCR,gene sequencing and immunohistochemical analysis shows that the placenta in the second trimester of pregnancy synthesize and secret AFP;3.The level of AFP mRNA in the elevated second trimester placental AFP complicated with adverse pregnancy outcomes group was significantly higher than that in the other two groups(P<0.0001).The relative expression of AFP mRNA and protein level in fetal liver were statistically different among the three groups(P<0.05),and the elevated second trimester placental AFP complicated with adverse pregnancy outcomes group was significantly lower than the other two groups.The level of ?-HCG mRNA in the elevated?-HCG complicated with adverse pregnancy outcomes group was significantly higher than that in the other two groups(P=0.0372),and the protein level in the healthy group was significantly higher than in the experimental group 1 and 2(P=0.0029).There was no difference in foetal membranes AFP and ?-HCG between the elevated level complicated with adverse pregnancy outcomes group and the healthy group and the normal level complcated with adverse pregnancy outcomes group(P>0.05).4.The results analyzed by the present samples showed that interleukin-1,interleukin-6,interleukin-10 and TNF-? in the placentas was not significantly different among the health group,elevated maternal serum AFP or/and ?-HCG complicated with adverse pregnancy outcomes group and the mormal level of maternal serum AFP and ?-HCG complicated with adverse pregnancy outcomes group(P>0.05).Conclusions:In this study,the expression of AFP in placenta,liver and fetal membranes were measured and quantified by cell morphology,protein and gene levels,and the lack in this field at home and abroad was provided,and theoretical reference for clinical diagnosis and treatment was suggested.However,gene regulation and specific pathogenesis resulting in the differences in the level of AFP in the three groups will be further studied.Based on those findings,we will further investigate the gene regulation by which AFP in the placenta express in different levels...
