Objective: To assess the effect of exercise on Anti-Mullerian Hormone and AMHRII in female rats with PCOS.Methods: 32 20-day-old female SD rats(66? ±? 14 g)were randomly divided into four groups: the control group,the control exercise(CE)group,the PCOS group and the PCOS exercise(PE)group.The exercise program was to swim with 120 min/d,6 times per week,for 15 days.The rat estrus cycle was observed via vaginal smears.The sex hormones,GnRH hormones and AMH in serum were detected by ELISA.The histological changes in ovarian tissues were observed with HE staining,and the expression of P450 aromatase and AMHRII in ovarian tissues were detected by immunohistochemistry.ANOVA and LSD test were used for data analysis.Results:Compared to Control group,CE group had no significant difference in estrous cycle?Ovary morphology,and P450 aromatase and AMHRII in ovarian tissues expression.Serum T and FSH tended to decline,while serum E2 and AMH tended to increase,but had no statistical significance(P>0.05).Also,compared to Control group,estrous cycle of PCOS group rested on anestrus for long time,and the ovaries were occupied by the early developmental follicles and atresia follicles,and there were cystic dilatation follicles.The serum E2(P<0.01)? T(P<0.01)?FSH(P<0.05)and AMH(P<0.01)were found increased markedly in PCOS group.P450 aromatase(P<0.05)and AMHRII(P<0.01)in ovarian tissues expression increased considerably.Compared to PCOS group,the estrous cycle and ovarian morphological parameters in PE group tended to be normal.And there was significantly lower serum T(P<0.01)?E2(P<0.05)and AMH(P<0.05)in PE group.The serum FSH?the expression of P450 aromatase and AMHRII in ovarian tissues showed a downward trend,but this was not statistically significant(P>0.05).Serum T?E2?AMH and FSH in PE group indicated an decreasing trend compared to the Control group but no significant differences were found(P>0.05),and the expression of P450 aromatase and AMHRII in ovarian tissues were the same(P>0.05),while the estrous cycle and ovarian morphological parameters in PE group tended to be normal.Conclusions:(1)Under the normal physiological conditions,regular exercises can decrease serum T,while cannot decrease serum AMH.(2)Our data suggests that DHEA injection to immature female rats can induce PCOS,which including hyperandrogenism,disorder of follicular development and anovulation;(3)Regular exercises can markedly decrease the serum AMH and T,decrease the expression of AMHRII in ovarian tissues,and restore to normal estrous cycle and ovarian morphology,which indicates that exercise mayimprove symptoms of polycystic ovary syndrome and ovarian hormones in women with abnormal ovarian function.(4)The mechanism may be: under the HPOA's normal control,regular exercises can decrease the expression of AMHRII via decrease serum AMH,and improve P450 aromatase activity,and thus facilitates serum T transform to serum E2.At the same time,the increased E2 strengthens the HPOA's feedback,therefore the release of LH decreased,and the serum T decreased.Thereby,the hyperandrogenism,dominant follicle development and reproductive functions have been improved.
