objective:Based on the preliminary study on the mechanism of aquaporin in the treatment of secretory otitis media(SOM)in mice by Qingqiao Capsule,a hospital preparation,this paper tries to provide a new theoretical basis for the treatment of secretory otitis media by Qingqiao Capsule,and to explore the effect of Qingqiao Capsule on the expression level of AQP1,4,5 in peripheral blood and middle ear mucosa of SOM mice.Proteomic mechanism of Qingqiao Capsule in the treatment of secretory otitis media.Methods : Eighty healthy mice with good auricle reflex and no history of noise exposure were randomly divided into healthy control group(10 mice),experimental group(35 mice)and blank control group(35 mice).he experimental group and blank control mice were sensitized by intraperitoneal injection of ovalbumin on the first and eighth days respectively.On the fifteenth day,ovalbumin was stimulated by intratympanic injection.The liquid level or liquid-gas level in tympanic chamber was observed within three days,which showed that the mouse model of secretory otitis media was successfully prepared and the unqualified mice in each group were eliminated.The experimental group was given 0.5 ml(0.02 mg/kg)of Qingqiao capsule suspension twice a day.The blank control group was given the same amount of saline by gavage.Two mice were randomly selected from the experimental group and the blank control group on d1,3,5,7 and 9,respectively,for heart blood collection and rapid scalp scraping of middle ear mucosa.Bilateral middle ear mucosa was scraped by rapid scalpel-off.The serum was centrifuged at high speed.The supernatant was obtained by high-speed centrifugation.The middle ear mucosa was decomposed at freezing point and then the supernatant was obtained by high-speed centrifugation.The contents of aquaporin AQP1,4,5 in peripheral serum and middle ear mucosa were determined by ELLSA in each group.Quantitative detection and statistical analysis?Statistical methods: SPSS 17.00 statistical software package was used to analyze the data.The measurement data were expressed as(+s).The comparison between groups was performed with two independent samples t-test,and the difference was statistically significant with P<0.05.Results:(1)The expression of AQP1 and 5 in serum of SOM mice in experimental group was significantly higher than that in blank control group(P < 0.01),the difference was statistically significant,while the expression of AQP4 was significantly lower than that in blank control group(P > 0.05);(2)The expression of AQP1 and 5 in ear mucosal lysate of SOM mice in experimental group was significantly higher than that in blank control group(P < 0.01),and the expression of AQP4 was significantly higher than that in blank control group(P < 0.01).There was no significant difference(P > 0.05).(3)The expression levels of AQP1,5 and AQP1,4 in SOM mice serum and middle ear mucosa were significantly higher than those in blank control group,while the expression levels of AQP5 in middle ear mucosa were significantly lower than those in blank control group,and there was no significant difference between the expression levels of AQP4 in serum and blank control group.Conclusion:(1)Qingqiao Capsule can significantly increase the expression of AQP1,5 in serum of SOM mice,increase the expression level of AQP1,5 in middle ear mucosa of SOM mice,and accelerate the expression of AQP1,5 in serum and AQP1,4 in mucosa of SOM mice.However,the expression level of AQP4 in middle ear mucosa and serum of SOM mice is limited,and there is no significant intervention on AQP4 in middle ear mucosa.(2)It is suggested that Qingqiao Capsule may interfere with the distribution of middle ear mucosal fluid in secretory otitis media(SOM)by increasing the expression level of aquaporin AQP1,5 in serum and middle ear mucosa of SOM mice,improve the efficiency of water transport of AQP1,4 in middle ear mucosa,regulate the water balance of middle ear mucosa,thus promote the clearance of middle ear effusion,shorten the course of SOM and improve the secretory middle ear.The prognosis of inflammation has positive and obvious effect;(3)but Qingqiao Capsule has limited regulation effect on aquaporin AQP 4 in middle ear mucosa and peripheral blood of mice.It may down-regulate the expression of AQP 5,indicating that there may be a coordinated expression between AQP1,4,5 in middle ear mucosa and serum,and there may be a balance of expression and distribution among different subtypes,but how does Qingqiao Capsule interfere with AQP1,4,5How to coordinate the expression of aquaporins and their signaling pathways need further study.
