Objective:To observe the influence of Shu-Mu Acupoint Application of Rexiao Formula on Th17/Treg imbalance in mice with asthma from the perspective of external treatment of traditional Chinese medicine.Methods:The acute bronchial asthma attack model was copied by OVA.60 female BALB/C mice were randomly divided into 6 groups: normal control group,model group,Shu-Mu acupoint application of Rexiao formula group,Shu-Mu acupoint application of The placebo group,sham acupoint application of Rexiao formula group,and the positive control group,with 10 mice in each group.In addition to the normal control group,other groups using OVA manufacturing asthma model.Six hours before each OVA stimulation,the Shu-Mu acupoint application of Rexiao formula group was treated with hot croup patch,the Shu-Mu acupoint application of The placebo group was treated with hot croup patch,the sham acupoint application of Rexiao formula group was treated with hot croup patch,and the positive control group was treated with dexamethasone gavage.The behavior and weight of mice in each group were observed.Mice were killed within 24 hours after the final atomization,blood and lung tissue were taken.HE staining was used to detect the pathological changes in lung tissues;ELISA was used to detect the Ig E、IL-17 and IL-10 expression in serum.Immunohistochemistry was used to detect the protein expressions of RORγt and Foxp3,The correlation between the ratio and expression of IL-17、IL-10.Results:1.Behavioral changes in mice: Compared with the model group,the symptoms of snoring,wheezing,irritability,and other symptoms of the mice in the group of the hot stagnation and stagnation group were alleviated;Fang sham acupoint application group no significant difference.2.Changes in body weight of mice: Before the challenge,there was no significant difference in body weight between the groups(P>0.05).After14 days of challenge,compared with the model group,the other groups were elevated to a certain extent.Shu-Mu acupoint application of Rexiao formula group weight was significantly increased(P<0.05)3.HE staining was used to observe the pathological results of lung tissue: in the model group,there were more inflammatory cell infiltration in the bronchi,alveolar and perivascular,mucinous gland hyperplasia,airway epithelial rupture,inflammatory cell exudation in the interstitial and bronchial lumen.And secretions,some have mucus plug formation.Compared with the model group,the performance of the above pathology was significantly reduced in the group of the Shu-Mu acupoint application of Rexiao formula group;there was no significant difference between the Shu-Mu acupoint application of The placebo group and the sham acupoint application of Rexiao formula group.4.The serum levels of Ig E,IL-17 and IL-10 were detected by ELISA.Compared with the normal control group,serum Ig E levels in the model group were significantly increased(P<0.05).Compared with the model group,Shu-Mu acupoint application of The placebo group,and sham acupoint application of Rexiao formula group,the Ig E content of the Shu-Mu acupoint application of Rexiao formula group decreased significantly(P<0.05).Compared with the normal control group,IL-17 content in the model group was significantly increased(P<0.05),and IL-10 content was significantly decreased(P<0.05).Compared with the model group,Shu-Mu acupoint application of The placebo group,and sham acupoint application of Rexiao formula group,the IL-17 content of the Shu-Mu acupoint application of Rexiao formula group decreased significantly(P<0.05),IL-10 content.Significantly elevated(P <0.05).5.Immunohistochemistry was used to detect the expression of Th17 and Treg-specific transcription factors ROR γ t and Foxp3 in lung tissue:compared with the model group,Shu-Mu acupoint application of The placebo group,and sham acupoint application of Rexiao formula group,the expression of RORγt protein in the Shu-Mu acupoint application of Rexiao formula group was significantly decreased(P<0.05),the expression of Foxp3 protein was significantly increased(P<0.05),and the ratio of ROR γ t/Foxp3 protein expression was significantly decreased(P<0.05).Conclusion:1.Shu-Mu acupoint application of Rexiao formula can alleviate the acute attack symptoms of mice with asthma,reduce the infiltration of airway inflammatory cells,and thus alleviate the effect of asthma attack.2.Shu-Mu acupoint application of Rexiao formula may regulate the imbalance of Th17/Treg by reducing the expression of cytokine IL-17 and increase the level of IL-10 in serum.3.Shu-Mu acupoint application of Rexiao formula may regulate the imbalance of Th17/Treg by down-regulating the expression of RORγt and increasing the expression of Foxp3 in the lung tissues. |
