| Objective: By observing the scores change of symtoms and signs of using BiMinGan Prescription to treat AR patients with Lung-Spleen Deficiency and cold, and observing the behavior of AR rats、pathology of its nasal mucosa and the contents of IL-17、IL-10 in its serum, we aim at exploring the clinical efficacy and immune mechanism of BiMinGan Prescription in treating AR, and providing a new clinical thinking and objective basis for AR’s treatment.Methods:Clinical study:60 patients who meet the inclusion criteria are divided into Chinese Medicine group(BiMinGan Prescription Shuijian Fu) and Western Medicine group(Loratadine oral or Budesonide nasal spray added). Record the scores of their symptoms and signs before、2 weeks after and 4 weeks after the treatment. Experimental study:32 female SD rats are randomly divided into blank group、model group、Chinese Medicine group (BiMinGan group) and control group(Loratadine group). Apart from the blank group, other groups establish AR rats model with OVA, and after the model is established, blank group and model group are intragastic infusion with normal saline, Chinese Medicine group with water extrat of BiMinGan Prescription,control group with Loratadine water solution. Observe the scores of rats symptoms 7 days、14 days and 28 days after administration. Detect cytokine of IL-17、IL-10 with ELISA after the 28 days administration, and kill the rats and observe the pathologic nasal mucosa.Results:Clinical study:Scores of symptoms and signs of Chinese Medicine group and Western Medicine group decreased 2 weeks and 4 weeks after the treatment with the scores before the treatment which have statistically significant difference (P＜0.01). Scores of symptoms and signs of Chinese Medicine group have no difference with the Western Medicine group 2 weeks after the treatment (P>0.05) and statistically difference 4 weeks after(P<0.05).2 weeks after the treatment, the obvious effective rate and the total effective rate of Chinese Medicine group are 36.7% and 96.7%, and those of Western Medicine group are 26.7% and 90%, and they have no statistically difference (P>0.05).4 weeks after the treatment, the obvious effective rate and the total effective rate of Chinese Medicine group are 83.3% and 100%, and those of Western Medicine group are 60% and 100%, and they have statistically difference (P＜0.05). Experimental study:7 days、14days、28 days after the administration, symptoms of rhinocnesmus、sneezing、thin nasal discharge of Chinese Medicine group and the control group have been more obviously relieved than the control group (P＜0.01).28 days after the administration, scores of symptoms of Chinese Medicine group have statistically difference with the control group (P＜0.05).Under the light microscope, epithelial cells of nasal mucosa of rats have degeneration、necrosis on different levels after the model is established, inflammatory exudate is seen in nose. While after the treatment, the inflammtory degree of nasal mucosa is relieved, and the Chinese Medicine group and control group have statistically difference with model group (P<0.05) and the degree of Chinese Medicine group and control group have no statistical difference (P>0.05). After the model is established, the contents of IL-10 of model group is obviously decreased and the IL-17 is increased, which have statistically significant difference with the blank group (P<0.01).After the treatment of BiMinGan Prescription or Loratadine, the contents of IL-10 is increased and IL-17 is decreased, Chinese Medicine group and control group have statistically difference with model group (P<0.05)Conclusion: It shows that BiMinGan Prescription is effective on treating AR according to its satisfactory treatment of significant efficiency and total effectiveness. Rats experiment shows that the possible mechanism of BiMinGan Presciption treating AR is influencing the contents of IL-17 and IL-10. |
