Objective: This experiment mainly observes the behavioral effects of drink on temulentia mice,and studies the pathological sections of liver,and related pathological indicators,as well as the memory-related substances to evaluate the sobering?protecting liver and memory effects of sober-up drinks.Methods: In the behavioral study,the experiment of Runner was to determine the first drop time and the numbers of drops for the mice within 3 minutes after drinking at 30 min,60 min,90 min,and 120 min,in order to study the coordination ability of the mouse body.The experiment of Water maze was to determine the time that spent on looking for the platform for the mice after drinking at 30 min,60 min,90 min,and 120 min to study their memory.The experiment of Pole-climbing was to determine the time and climbing ability of the mice from the top to the bottom of the rod after drinking at 30 min,60 min,90 min,and 120 min to study their body strength and coordination ability.The Balanced test was performed to determine the first drop time from the rope in 3 minutes after drinking at 30 min,60 min,90 min,and 120 min to study its balanced ability and body strength on the mice after drinking.The experiment performed HE staining on the liver and kidney of mice after drug administration,and observed the section under the light microscope,and evaluated the steatosis of liver,levels of inflammation,et al.And determining the contents of ALT,AST in the blood and the contents of SOD,CYP2E1 in the liver were to determine its protective effects on the liver of temulentia mice.This experiment determined the contents of Ach,DA,NE which were memory-related substances in the brain of mice to study the effects of sober-up drinks on memory.Results: The first drop time of model group was shorter than that of the negative group,and the number of drops was more(P<0.05)in the Runner experimental.The first drop time of most of the drug-administered groups were longer than that of the model group and the numbers of drops were less than that of the model group.During the test period,the number of drops of the positive control group,the middle-low dose group,the medium-high dose group,and the high dose group were less than the model group(P<0.05).The first drop time of the positive control group,the middle-low dose group,high dose group were longer than the model group(P<0.01 or P<0.05).The time of model group was longer than that of the negative group in the water maze experimental(P<0.05).The search time of most of the drug-administered groups was lower than that of the model group,especially,the low-dose group,the middle-dose group and the middle-high dose group were significantly different(P <0.01 or P <0.05).Compared with the negative group,the climbing time of model group was shorter than that of the negative group and the level of climbing rod was higher in the experimental of pole-climbing(P<0.01).The score of positive group,middle-low dose group,and high dose group were lower than the model group(P<0.01 or P<0.05);the climbing time of the low dose group and the high dose group were higher than that of the model group(P <0.01 or P <0.05).The first drop time of model group was shorter than that of the negative group in the balanced experimental(P<0.05),and the drop time of the positive group,the middle dose group and the high dose group were longer than that of the model group(P<0.01 or P<0.05).Compared with the negative group,the liver slices of the model group showed that the liver cell was swelling,the liver with macrovesicular steatosis,the central venous was dilatation,the inflammatory cell was infiltration and the liver with punctate necrosis.Renal tubular water degeneration occurred in the kidney.In the drug-administered group,the vacuolar degeneration was less,the cell swelling and inflammation were lighter,and the kidney was almost free of renal tubular water degeneration.The values of ALT,AST and pathological sections in the model group were higher than the negative group(P<0.01 or P<0.05),as well as the values of SOD were lower(P<0.05),and the values of CYP2E1 were smaller(P<0.05).The values of AST and ALT of all drug-administered groups were lower than those of the model group.The values of ALT of the middle-low-dose group,the middle-dose group and the medium-high dose group were significantly different(P<0.01 or P<0.05).The values of AST of the groups,which excepted the high dose group,were significantly different(P<0.01 or P<0.05),while the values of SOD of the high dose group were significantly higher(P<0.05).The values of CYP2E1 of positive group were higher than the model group(P<0.05).The scores of the pathological section were lower than the model group(P<0.05).The values of Ach of the model group were lower than that of the negative group(P<0.05).The values of DA and NE of the positive group,the low dose group,the middle-high dose group and the high dose group were higher than those of the model group(P<0.01 or P<0.05).The values of Ach of the positive group and low dose group were higher than that of the model group(P<0.05).Conclusion: The models of temulentia mice are made successfully in the above experiments.The sober-up drink can improve the body strength,balance ability,coordination ability and memory ability of the drunken mice to reach the effect of sober-up.It can also protect the liver and improve the memory ability of the temulentia mouse.
