Objective:The method of model that let the rats drink different low concentration of alcohol beverage freely, establishment of chronic alcoholism which is simple and similar to the ideals of chronic alcoholism of humans.Methods:(1)Eighty general level adult Wistar rats (370±30)g were randomized into control groupl、6%alcohol groupl、8%alcohol groupland12%alcohol groupl, control group2、6%alcohol group2、8%alcohol group2and12%alcohol group2,each group was6.(2)Use gradually increasing the alcoholic strength by the establishment of chronic alcoholism in rats model of free drinks. Model groups of rats free drink the concentration of alcoholic beverages every day, do not give water, control groups of rats given tap water, to eat food casually. Models were carried out in two phases, that is3month and4month.(3)Through Morris water maze to test injury of learning and memory, Gas chromatographic method to detect blood alcohol concentration, using optical microscopy and electron microscopy observation of rat pathological changes in the brain and liver after alcohol poisoning respectively.Result:(1)Free drinking alcohol at the same time, the alcohol content intake in the body was the most in12%alcohol group,8%alcohol group followed by the alcohol group of12%,6%alcohol group at least. Compared with the control group, the model groups of rats of weight gained slowly,12%alcohol group is particularly significant.(2)The degree of injury for learning and memory was12%alcohol group most significantly, the changes in6%alcohol group and8%alcohol group was not significant.(3)After modeled3-and4-month, the blood alcohol concentration in model groups of rats was almost between150-200mg/100ml,12%alcohol group was the most high,8%alcohol group followed by the alcohol group of12%,6%alcohol group at least.(4)Through the optical microscopic to observe, after3months although the pathological changes in the liver, but did not appear in the brain. Through the transmission electron microscope to observe the frontal cortex, cerebellum and hippocampus of Synapse gap thickness were thin after modeled4months in the models group, the number of synaptic was reduced. the number of synaptic vesicles were increased, the synaptic gap was narrowed, the synaptic thick membrane of dense electron density was reduced, the bilayer membrane of mitochondria and crest structure in the synaptic were not clear or destroyed, the crest was fractured、shorter and reduced.Conclusion:(1)After Sand4months the model groups of rats can be both established the chronic alcoholism models of rats in free drink alcoholic beverages at low concentration. But after3month it was only appeared behavioral changes and pathological changes of liver, the pathological changes of brain was appeared after4months,12%models of alcohol group were most available for reference.(2)The method for determination of concentration of alcohol in the blood, can provide an accurate alcohol poisoning in rats.(3)After4months the model groups of rats can be appeared the ultrastructure changes of brain and liver.
