ObjectiveTo observe the therapeutic effect of honeysuckle granules on acne in rabbit ear and to its regulatory effect on NF-?B signaling pathway.MethodsCoal tar was applied on the medial side of rabbit ear once a day for 14 days until acne was formed.After successful modeling,the Male rabbits were randomly divided into blank group,isodimension A acid group,honeysuckle granules high dose group,honeysuckle granules middle dose group,honeysuckle granules low-dose group and model group.After 14 days of treatment,Detection of NF-?Bp65,IKK-?/? in male rabbits ear organization by western blot.Determination of IL-1? and TNF-? in Serum of male rabbits by ELISA.The changes of medial skin in rabbit ear group were observed by HE staining under microscope and naked eyes during the treatment cycle.ResultsThe serum levels of TNF-? and IL-1? in male rabbits in the model group were significantly higher than those in the blank group(P<0.01);The contents of IL-1? and TNF-? in the Honeysuckle granules medium dose group,Honeysuckle granules high dose group and group of isotretinoin were significantly lower than those in the model group(P<0.01).The content of serum TNF-? in the low dose group of honeysuckle granules was lower than that in the model group(P<0.05);the content of IL-1? was significantly lower than that of the model group(P<0.01);The expression of NF-kappa Bp65,IKK-? and IKK-? protein in the model group was higher than that in the blank group(P<0.01).Compared with the model group,the expression of NF-kappa Bp65,IKK-? and IKK-? in the high dose group,middle dose group and isotretinic acid group were significantly down-regulated(P<0.01).The expression of NF-kappa Bp65 protein was decreased in the low dose group of honeysuckle(P<0.05).The expression of IKK-? and IKK-? protein was down regulated in the low dose group of honeysuckle(P<0.01).ConclusionFlos Lonicerae has a definite therapeutic effect on acne by regulating NF-?B signaling pathway to regulate intracellular NF-?Bp65,IKK-?,IKK-? protein concentration and serum TNF-? and IL-1? levels.
