Objective:The aim of this study was to apply ultra-high performance liquid chromatography-mass spectrometry(LC-MS)to metabolomics metabolomics studies in urine samples from patients with acute coronary syndrome(ACS)and healthy controls to further explore acute crowns.Pathogenesis of pulse syndrome.Methods:This study included patients who underwent coronary angiography in the Department of Cardiology,People's Hospital of Xinjiang Autonomous Region from January 2017 to September 2018.There were 48 patients with acute coronary syndrome(35 patients with acute myocardial infarction,13 patients with unstable angina pectoris)and 12 healthy controls.Urine samples were separated by ultra-high performance liquid chromatography-mass spectrometry using principal component analysis(PCA),partial least squares discriminant analysis(PLS-DA analysis and orthogonal partial least squares discriminant analysis(OPLS)-DA)The variable projection importance value(VIP)in the model characterizes the difference endogenous between groups.Each group of characteristic metabolic pathways was screened and profiled for specific metabolic pathway-related metabolites.Results: There were significant differences in the urinary metabolite profiles between the ACS and the control group.The differential pathways were mainly:1.Control group and myocardial infarction group: Sphingolipid metabolism.2.Control group and angina group: Nitrogen metabolism,alanine,aspartate and glutamate metabolism,arginine and proline metabolism pathway(Arginine)And proline metabolism),Nicotinate and nicotinamide metabolism,Phenylalanine metabolism,Aminoacyl-tRNA biosynthesis.3.Myocardial group and angina group: Alanine,aspartate and glutamate metabolism,Purine metabolism,Nitrogen metabolism.20 differential biomarkers of 8 metabolic pathways were significantly up-regulated,including Phytosphingosine,Sphinganine,Calcium formate,L-glutamine,L-L-arginine,L-tyrosine,L-tryptophan,Serinyl-glutamate,aspartic acid(L-Aspartate),L-asparagine,Oxaloacetate,Succinate,Glutamate,Spermidine,Spermine,Maleic Acid Maleate,Tyrosine,Phenylalanine,L-histidine,Hypoxanthine.Conclusion: This study shows sphingolipids,nitrogen,alanine,aspartic acid and glutamic acid,arginine and proline,niacin and nicotinamide,phenylalanine,guanidine metabolic pathway,aminoacyl biosynthetic pathway Play an important role in the pathogenesis of ACS.Therefore,urology metabolomics has the potential to improve the diagnostic efficacy of ACS,making it more accurate and comprehensive for the diagnosis of ACS.
