Objective:To investigate the relationship between soluble low-density lipoprotein receptor 11(sLR11)gene rs641120 polymorphism and sLR11 level and type 2 diabetes mellitus(T2DM)with macrovascular disease in Yanbian area.Research object and method:237 patients with type 2 diabetes mellitus were treated with endocrine and hospitalization in yanbian University Hospital from December 2014 to December 2015.Among them.T2DM carotid atherosclerosis group(CAS group,133 cases),and no carotid atherosclerosis group(NCAS group,104 cases)Another normal control group(NGT group,76 cases,healthy people)was set up.Carotid IMT and plaque were measured using a color ultrasound system.SNP typing and sequencing were performed by the LDR-PCR method.The whole blood genomic DNA extraction kit extracts genomic DNA.sLR11 levels were determined using a monoclonal antibody detection sLR11 kit.Data analysis was performed using SPSS 24.0 statistical software.Results:1.The rs641120 locus of sLR11 gene was T>C in NGTr NCAS and CAS,and the genotype frequencies were CT>TT>CC,no statistical difference(P>0.05).At the same time,the sLR11 gene rs641120 allele and genotype frequencies of NGT group,NCAS and CAS group were analyzed under dominant and recessive patterns,and there was no statistical difference(P>0.05).2.The levels of BMI,SBP,DBP,FPG,TG and sLR11 were significantly higher in the CAS group and NCAS group than in the NGT group;and TC and LDL-C were significantly higher in the CAS group than in the NGT group(P<0.05)..3.The statistical results of rs641120 polymorphism genotypes in the relevant groups showed that the BMI was significantly higher in the CT group than in the TT group in the NCAS group;the SBP in the NCAS group was higher in the CC group than in the TT group,and in the C-C group and the CT group.In the TT group,the decreasing trend was observed.In the NGT group,the CC group was higher than the TT group,and it showed a decreasing trend in the CC group,CT group and TT group,with statistical difference(P<0.05).4.FPG,LDL-C is a risk factor for IMT thickening;HDL-C,rs641120 wild homozygous CC is a protective factor for IMT thickening,with statistical difference(P<0.05).5.Age.SBR FPG,TG and sLR11 levels were correlated with plaque formation,with statistical difference(P<0.05)..6.The P value of Age,BMI,SBP,FPG,TC,TG was<0.05,and the R values were 0.195,0.129,0.147,0.242,0.143,0.151,respectively.It is known that these factors are positively correlated with sLR11 level,and there is statistical difference(P<0.05).7.Age,BMI,FPG,TC P value<0.05,and OR value>1,we can see that these factors are risk factors for sLR11 level,there is statistical difference(P<0.05)Conclusion:1.The sLR11 gene rs641120 polymoiphism is not associated with T2DM.2.sLR11 gene rs641120 wild homozygous CC is a protective factor for preventing IMT thickening,and increases the occurrence of CAS when C?T mutation.3.The level of sLRll is correlated with age,weight blood pressure,blood glucose and blood lipids,and may be involved in plaque formation in T2DM.
