Objective:To investigate the correlation between soluble low density lipoprotein receptor 11(sLR11)gene rs2070045 single nucleotide polymorphism(SNP),sLR11 levels and carotid atherosclerosis in patients of type 2 diabetes mellitus(T2DM)belonging to two ethnic groups(Han and Korean)within Yanbian Region.Methods:This study was conducted on 237 patients with type 2 diabetes mellitus(T2DM)who received outpatient and inpatient treatment at the Department of Endocrinology,Yanbian University Hopital between June 2016 and June 2017.Based on the carotid IMT values,the T2DM group were divided into carotid atherosclerosis group(CAS group,n=133,Carotid IMT≄1.0mm with plaque),and non-carotid atherosclerosis group(NCAS group,n=104,IMT<1.0mm).The normal control group(NGT group)consisted of 76 healthy individuals.Color Ultrasound was used to measure Carotid IMT and to determine presence of plaque.Genomic DNA was extracted from whole blood.SNP typing and sequencing was done by PCR-LDR method.Serum SLR11 levels were measured by using a monoclonal antibody detection kit.The risk factors of carotid atherosclerosis were screened using Logistic regression analysis,with P<0.05 indicating statistical significance.Data analysis was performed using SPSS25.0 software.Results:1.There was no statistical difference in genotype and allele frequency distribution between NGT and NCAS,NGT and CAS,NCAS and CAS group,(P>0.05).2.The sLR11 gene rs2070045 allele and genotype frequencies of NGT,NCAS and CAS group were analyzed under dominant and recessive patterns,and there was no statistical difference(P>0.05).3.Statistical analysis of GG,GT and TT genotypes in the NGT,NCAS and CAS groups show that LDL-C is higher in the TT group than in the GG and GT groups in the CAS group(P<0.05).4.BMI,SBP,DBP,FPG,and sLR11 level were significantly higher in the CAS and NCAS groups than in the NGT group;and TC and LDL-C were significantly higher in the CAS group than in the NGT group(P<0.05).5.Sex(Male),FPG,HDL-C,LDL-C,atherosclerotic plaque,smoking,insulin supplementation,medication for hypertension,medication for dyslipidemia have statistically significant correlation with IMT thickening(P<0.05).FPG,LDL-C have OR value>1,which indicates these are risk factors for IMT thickening,whereas HDL-C and rs2070045GG genotype have OR value<1,which indicates these are protective factors for IMT thickening(P<0.05).6.Age,SBP,FPG,TG,sLR11,thickened IMT,renal dysfunction,insulin supplementation,medication for hypertension,medication for dyslipidemia and rs2070045GG genotype have statistically significant correlation with atherosclerotic plaques(P<0.05).Age,SBP and FPG have OR value>1,which indicates these are risk factors for atherosclerotic plaques,whereas rs2070045GG genotype has OR value<1,which indicates this is a protective factor for atherosclerotic plaques(P<0.05).7.Sex,age,BMI,SBP,FPG,TC,TG,smoking and oral hypoglycemics have statistically significant correlation with sLR11 levels.Age,BMI and FPG have OR value>1,which indicates these are risk factors for sLR11 levels(P<0.05).Conclusion:1.The sLR11 gene rs2070045 polymorphism showed no significant correlation with T2DM.2.The sLR11 gene rs2070045 homozygote TT is a risk factor for carotid IMT thickening and development of atherosclerotic plaques.3.sLR11 levels showed significant correlation with age,BMI and FPG,and may be involved in development of carotid atherosclerosis in T2DM patients.
