| Objective This study observed the effects of Tongfeng Kangning Formula on serum uric acid,XOD,ADA,IL-6,IL-8,CRP and protein expression of the signaling pathway of the toll-like receptor 2(TLR2)/myeloid differentiation factor(MYD)88 in ankle joint synovial tissue of acute gouty arthritis(AGA)rats..Method After 1 weeks of adaptive feeding,48 Wistar rats were selected and 8 were randomly selected as the normal control group and were given routine feeding.The rest 40 were divided into model group,colchicine group,gout Tongfeng Kangning Formula low,middle and high dose group randomly,and 8 rats in each group.Combined xanthine(HX)gavage and urate inhibitor potassium oxazine(OAPS)were given to the model group and each drug group every morning for 7d.On the fifth day rats after administration were measured right hind ankle diameter,and uric acid sodium solution was injected into the lateral ankle joint cavity,establish the model of acute arthritis,respectively after injection of 1h,8h,12 h,24h,48 h of rats measured ankle diameter,observe ankle joint swelling degree.The intervention drugs were given to the stomach every afternoon,and the normal and model groups were given aseptic saline.After 1 weeks of treatment,rats by intraperitoneal injection of chloral hydrate anesthesia,abdominal aortic blood,separation of serum,for the detection of serum uric acid and XOD?ADA?IL-6?IL-8?CRP,remove the right hind ankle samples,10% Formaldehyde Solution fixed,decalcified,dehydrated decalcifying agent.Transparent,paraffin embedding,sectioning and staining.The pathology of the ankle joint of rats was observed under light microscope and the proteion expression of TLR2 and TLR4 and MyD88 was determined dy immunohistochemistry.Result 1 rats showed that compared with the control group,the level of serum uric acid and xanthine oxidasea and adenosine deaminase increased significantly in the model group(P<0.01),which proved that the model was successful.Tongfeng Kangning Formula is low,medium and high dose group and colchicine group.The serum uric acid and xanthine oxidase and adenosine deaminase were compared with those of model group were decreased(P<0.01).2 rats showed that serum interleukin-6 and interleukin-8 levels in model group were significantly higher than those in control group(P<0.05).Compared with the model group,the levels of serum interleukin-6 and interleukin-8 in each group were decreased(P<0.05 or P<0.01).rats showed that serum C-reactive protein in model group were significantly higher than those in control group(P<0.05).Compared with the model group,the levels of serum C-reactive protein in each group were decreased(P<0.05 or P<0.01).3 the pathological changes of the ankle joint of rats in each group: the synovial membrane structure of the control group was clear,the synoviocytes were arranged neatly,no hyperplasia,no inflammatory cell infiltration.In the model group,the surface of the synovial membrane was rough and the cell proliferation was obvious,and a large number of inflammatory cells were infiltrated.Colchicine,Tongfeng Kangning Formula low,medium and high dose group of synovial cell hyperplasia degree compared with the model group,were reduced,inflammatory cell infiltration is small scattered.4 compared with the normal control group,the expression of TLR2 and TLR4 and MyD88 in synovial tissue of joint in the model control group were all increased,indicating the significant difference(P<0.05).the colchicine group and Tongfeng Kangning Formula groups compared to the model group,the protein expression of TLR2 and TLR4 and MyD88 in synovial tissue decreased significantly(P<0.05),the join sewlling in test join decreased significantly.Conclusion 1 Tongfeng Kangning Formula can inhibit the secretion of xanthine oxidase and adenosine deaminase,reducing uric acid generation.2 Tongfeng Kangning Formula can significantly alleviate gout rats ankle swelling,inhibiting the expression of inflammatory factor interleukin-6 and interleukin-8,C-reactive protein.3 Tongfeng Kangning Formula can promote the repair of synovial tissue of rat ankle.4 Tongfeng Kangning Formula can inhibit the protein expression of TLR2 and TLR4 and MyD88 in synovial tissue of acute gouty arthritis(AGA)rats.. |
